目的 对肠道病毒71型(EV71)杭州株H3-TY的基因特征进行分析并探讨其用于疫苗开发的可能性.方法 利用RT-PCR方法扩增EV71 H3-TY株全基因组并克隆入质粒载体,对克隆基因进行序列测定并拼接为全长序列.其后利用Lasergene及MEGA软件对所获序列与11株中国大陆C4亚型病毒株、中国台湾B型代表株TW和国际标准株(BrCr)进行同源性比较,并进行进化树分析.结果 EV71 H3-TY株属于C4亚型,与其他中国大陆流行株在核苷酸水平的同源性介于94.0% ~99.0%,氨基酸水平的同源性均高于97%.H3-TY主要抗原位点SP70与其他毒株完全相同,SP55除了与BJ08株及BrCr株略有不同外,与其他毒株也均相同.结论 EV71 H3-TY株与其他国内流行株高度同源,主要抗原位点一致,可用于疫苗开发研究.

Objective To investigate genetic characteristics of enterovirus 71(EV71) Hangzhou isolate H3TY and explore the possibility of vaccine development.Methods The whole genome of EV71 strain H3-TY was amplified by RT-PCR and cloned into plasmid for sequencing.The genome sequence was thus obtained by splicing sequences from individual clones and was taken homology and phylogenetic analysis with C4,TW and BrCr strains by Lasergene and MEGA.Results The strain H3-TY belonged to subtype C4.When compared to other domestic strains,the homology value of nucleotide was between 94.0% and 99.0%,and the homology value of amino acid was over 97%.H3-TY major antigen site SP70 was the same as other strains,and SP55 was also the same as other strains,but slightly different fiom the BJ08 strain and BrCr strain.Conclusions EV71 strain H3-TY has a high level of homology with other domestic strains and shares the same major antigen site,which could be potentially useful for development of EV71 vaccine.
