多项全基因组关联分析认为,IL-28B基因多态性与聚乙二醇IFN-α联合利巴韦林治疗慢性丙型肝炎效果之间存在密切联系,同时还发现IL-28B基因多态性与HCV自发清除、肝脏脂肪变性、生化指标改变有密切关联.IL-28B优势基因型的丙型肝炎患者,肝脏表达IL-28B及干扰素刺激基因的基线水平较低,给予外源性IFN-α后,IL-28B诱导增强,能够更有效地清除病毒.此文总结了目前IL-28B在HCV感染及治疗应答方面的作用.

A series of genome-wide association studies have identified that there is an intimate connection between IL-28B polymorphisms and the outcome of pegylated-IFN-α plus ribavirin combination therapy in chronic hepatitis C.At the same time,IL-28B has been reported to associate with HCV spontaneous clearance,hepatic steatosis and biochemical changes.In patients with favorable IL-28B genotype,hepatic expression levels of IL-28B and interferon-stimulated genes are low at baseline; after IFN-α administration,IL-28B level enhances,resulting in more effective clearance of HCV.This review summarizes the recent understanding of IL-28B roles in HCV infection and response to therapy.
