目的 探讨妊娠特异性糖蛋白(PSG)对Balb/c小鼠脾脏来源的树突状细胞(DC)的免疫调节作用.方法 无菌分离Balb/c小鼠脾脏单个核细胞,rmGM-CSF、rmIL-4诱导分化为DC,同时施加0.2 g/L PSG作用7d,并设对照组,流式细胞仪检测DC表面标志和抗原摄取能力,MTT法检测其同种异基因淋巴细胞刺激能力.结果 与对照组相比较,DC表面标志MHC-Ⅱ类分子、CD80、CD86和CD40的表达显著低于对照组(t=2.52,t =3.24,=2.74,t =2.58,P<0.01),DC抗原摄取能力亦明显降低(t=3.15,P<0.01),刺激细胞:反应细胞比例为1∶10和1∶20的PSG作用后的DC刺激同种T淋巴细胞增殖的能力明显降低(t=3.12,t =2.76,P<0.01),比例为1∶50和1∶100的PSG组T淋巴细胞增殖能力也有降低(t=2.34,t =2.47,P<0.05).结论 PSG可下调Balb/c小鼠脾脏DC的免疫功能,可能与妊娠免疫抑制有关.

Objective To investigate the regulation of pregnant specific glycoprotein (PSG) on dendritic cells derived from spleen of Balb/c mice.Methods Spleen mononuclear cells of Balb/c isolated under sterile conditions were induced to differentiate into dendritic cells with rmGM-CSF and rmIL-4,Cells were treated with 0.2 g/L PSG for 7 days.Surface markers and the capacity of antigen uptaking of dendritic cells were analyzed with flow cytometry.The capacity of DCs in stimulating allogene lymphocytes was detected by methyl thiazolyl tetrazolium (MTT) assay.Results Compared with control,the dendritic cells treated with PSG expressed lower MHC-Ⅱ,CD80,CD86 and CD40 (t =2.52,t =3.24,t =2.74,t =2.58,P <0.01).The competence of antigen uptaking decreased differently (t =3.24 and P < 0.01).The stimulating capability of allogeneic T lymphocytes that the ratio of stimulating cell and responding cell was 1 ∶ 10 and 1 ∶ 20 decreased apparently(t =3.12,t =2.76,P <0.01) and that of 1∶50 and 1∶ 100 also decreased(t =2.34,t =2.47,P <0.05).Conclusion PSG can down-regulate the immune function of the dendritic cells derived from spleen of Balb/c mice.This negative effective effect of PSG might be associated with pregnant immune suppression.
