目的 研究神经干细胞(NSCs)移植联合氨基甲酰促红细胞生成素(CEPO)对新生脑瘫大鼠脑损伤的保护作用.方法 从孕14~16天大鼠获得神经干细胞,经过培养并传代后备用.利用7日龄wistar大鼠制作脑瘫模型,给予CEPO腹腔注射和神经干细胞移植,对神经干细胞移植后的分化、凋亡情况,免疫分子在移植前后的改变,大鼠神经系统功能检测等各项指标进行评估,并与单一神经干细胞移植、CEPO腹腔注射效果进行比较.结果 缺氧缺血脑损伤后,IFN-γ和IL-1β的浓度显著提高(P<0.05),MHC的含量增加,大鼠学习能力下降(P<0.05)..联合应用CEPO和神经干细胞移植可使移植细胞存活率提高并向神经元分化,降低血中IFN-γ和IL-1β水平,调节MHC抗原的表达,进而明显改善大鼠的神经运动功能.结论 应用CEPO可促进移植细胞存活,调节免疫分子的分泌,改善细胞存活的环境,对移植细胞具有明显的保护作用.
目的 研究神經榦細胞(NSCs)移植聯閤氨基甲酰促紅細胞生成素(CEPO)對新生腦癱大鼠腦損傷的保護作用.方法 從孕14~16天大鼠穫得神經榦細胞,經過培養併傳代後備用.利用7日齡wistar大鼠製作腦癱模型,給予CEPO腹腔註射和神經榦細胞移植,對神經榦細胞移植後的分化、凋亡情況,免疫分子在移植前後的改變,大鼠神經繫統功能檢測等各項指標進行評估,併與單一神經榦細胞移植、CEPO腹腔註射效果進行比較.結果 缺氧缺血腦損傷後,IFN-γ和IL-1β的濃度顯著提高(P<0.05),MHC的含量增加,大鼠學習能力下降(P<0.05)..聯閤應用CEPO和神經榦細胞移植可使移植細胞存活率提高併嚮神經元分化,降低血中IFN-γ和IL-1β水平,調節MHC抗原的錶達,進而明顯改善大鼠的神經運動功能.結論 應用CEPO可促進移植細胞存活,調節免疫分子的分泌,改善細胞存活的環境,對移植細胞具有明顯的保護作用.
목적 연구신경간세포(NSCs)이식연합안기갑선촉홍세포생성소(CEPO)대신생뇌탄대서뇌손상적보호작용.방법 종잉14~16천대서획득신경간세포,경과배양병전대후비용.이용7일령wistar대서제작뇌탄모형,급여CEPO복강주사화신경간세포이식,대신경간세포이식후적분화、조망정황,면역분자재이식전후적개변,대서신경계통공능검측등각항지표진행평고,병여단일신경간세포이식、CEPO복강주사효과진행비교.결과 결양결혈뇌손상후,IFN-γ화IL-1β적농도현저제고(P<0.05),MHC적함량증가,대서학습능력하강(P<0.05)..연합응용CEPO화신경간세포이식가사이식세포존활솔제고병향신경원분화,강저혈중IFN-γ화IL-1β수평,조절MHC항원적표체,진이명현개선대서적신경운동공능.결론 응용CEPO가촉진이식세포존활,조절면역분자적분비,개선세포존활적배경,대이식세포구유명현적보호작용.
Objective To investigate effects of combined transplantation of neural stem cells and carbamylated erythropoietin (CEPO) on neonatal rats with cerebral palsy,and discuss the underlying mechanisms of treating central nervous system injuries.Methods We utilized neonatal rats with cerebral palsy,at different time points,CEPO was given by intraperitoneal injection,or NSCs were transplanted in ventricle,or NSCs transplantation was combined with CEPO.The differentiation and apoptosis of NSCs,and the changing of inflammation cytokines and immune molecules were investigated.Neurological functions were assessed and compared with single NSCs transplantation or CEPO injection to explore the possible better way of treatment.Results The NSCs were successfully cultured.In neonatal rats with cerebral palsy,result of ELISA showed that IFN-γand IL-1 β were increased in serum,while CEPO could decrease their levels.In transplanted ones,both MHC-Ⅰ and MHC-Ⅱ were increased,and down-regulated when CEPO was administered.Moreover,the combination of CEPO injection and NSCs transplantation could accelerate neurological recovery and the survival and differentiation to neurons of transplanted cells.Conclusions In neonatal rats with cerebral palsy,combination of CEPO and NSCs transplantation promoted survival and differentiation of NSCs,and decreased the level of IFN-γ and IL-1 β in serum,and regulated MHC expressions,and then promoted neurological recovery.
