目的 筛选曲古菌素A作用胃癌细胞系SGC-7901后明显表达下调的基因,检测其在临床胃癌组织中的表达水平.方法 采用基因芯片技术,检测胃癌细胞系SGC-7901经过曲古菌素A干预后基因表达的变化,筛选明显表达下调的基因,应用免疫组化检测候选分子在胃癌及癌旁正常组织中的表达.结果 胃癌细胞系SGC-7901经75 ng/mL曲古菌素A作用48 h后,Dickkopf related protein-1(DKK1)基因表达下调,差异具有统计学意义(P=0.037 0).免疫组化显示胃腺癌组织中DKK1基因表达明显高于周围正常胃组织,二者表达差异具有统计学意义(P =0.000).结论 DKK1基因在曲古菌素A作用胃癌细胞系SGC-7901后表达下调.DKK1基因在胃腺癌组织中表达增高.

Objective We investigated that the down-regulated genes were involved in Trichostatin A treatment in gastric cancer SGC-7901 cells.The expression of the candidate gene was detected in clinical gastricadenocarcinoma cases.Methods The changes of gene expression were detected in gastric cancer cell line SGC-7901 after Trichostatin A intervention by using gene chip technology.The down-regulated genes were screened and the expression was tested in gastric adenocarcinoma tissues by immunohistochemical analysis.Results Gastric cancer SGC-7901 cells with 75 ng/mL after 48 h of Trichostatin A treatment showed down-regulation of Dickkopf related protein-1 (DKK 1) gene expression.The differences of DKK1 expression were significant (P =0.037 0).Immunohistochemistry result showed that the expression of DKK1 in gastric adenocarcinoma was significantly higher than that in the surrounding normal tissues (P =0.000).Conclusion DKK 1 expression was decreased in gastric cancer SGC-7901 cells after Trichostatin A treatment.DKK1 is overexpressed in gastric adenocarcinoma tissue.
