目的 探讨冬虫夏草对大鼠慢性阻塞性肺疾病(COPD)模型还原型谷胱甘肽(GSH)-氧化型谷胱甘肽(GSSG)失衡的干预作用及对Th1-Th2型细胞因子的影响.方法 18只SD大鼠完全随机分为COPD对照组、冬虫夏草干预组和N-乙酰半胱氨酸(NAC)干预组(每组各6只),分别在COPD模型制作中胃饲生理盐水、冬虫夏草和NAC,观察大鼠肺组织病理改变.检测冬虫夏草和NAC干预后支气管肺泡灌洗液(BALF)中Th1-Th2型细胞因子和巨噬细胞中GSH和GSH/GSSG变化.结果 与COPD对照组(32±13)相比,冬虫夏草干预组和NAC干预组的平均肺泡计数增加(49±10,52±14,P<0.05).冬虫夏草干预组和NAC干预组BALF中巨噬细胞GSH水平和GSH-GSSG较COPD对照组升高(t=3.06,t=3.24; t=2.36,t=2.82;均P<0.05).冬虫夏草干预组和NAC干预组BALF上清液中Th1型细胞因子IFN-γ水平较COPD对照组升高(t=2.34,t=2.32,P<0.05).结论 冬虫夏草可能通过提高COPD模型巨噬细胞内GSH水平,补充抗氧化水平而达到纠正Th1-Th2失衡的免疫调节.

목적 탐토동충하초대대서만성조새성폐질병(COPD)모형환원형곡광감태(GSH)-양화형곡광감태(GSSG)실형적간예작용급대Th1-Th2형세포인자적영향.방법 18지SD대서완전수궤분위COPD대조조、동충하초간예조화N-을선반광안산(NAC)간예조(매조각6지),분별재COPD모형제작중위사생리염수、동충하초화NAC,관찰대서폐조직병리개변.검측동충하초화NAC간예후지기관폐포관세액(BALF)중Th1-Th2형세포인자화거서세포중GSH화GSH/GSSG변화.결과 여COPD대조조(32±13)상비,동충하초간예조화NAC간예조적평균폐포계수증가(49±10,52±14,P<0.05).동충하초간예조화NAC간예조BALF중거서세포GSH수평화GSH-GSSG교COPD대조조승고(t=3.06,t=3.24; t=2.36,t=2.82;균P<0.05).동충하초간예조화NAC간예조BALF상청액중Th1형세포인자IFN-γ수평교COPD대조조승고(t=2.34,t=2.32,P<0.05).결론 동충하초가능통과제고COPD모형거서세포내GSH수평,보충항양화수평이체도규정Th1-Th2실형적면역조절.
Objective To investigate the effects of Cordyceps sinensis (C.sinensis) on reduced/oxidized glutathione imbalance and Th 1/Th2 cytokines in rat models with chronic obstructive pulmonary disease (COPD).Methods A total of 18 Sprague-Dawley male rats was randomly assigned to receive normal saline(control group,n=6),C.sinensis(C.sinensis intervention group,n=6) or N-acetylcysteine(NAC intervention group,n=6) feeding for determination of pathological changes in lung tissues.Bronchoalveolar lavage fluid(BALF) was collected to determine the changes in Th1/Th2 cytokines and variation in reduced/oxidized glutathione within macrophages after interventions of C.sinensis and NAC.Results Both C.sinensis and NAC resulted in increased alveolar cell count and the ratio of reduced to oxidized glutathione within macrophages obtained from BALF as compared with the counterpart controls(49±10 and 52±14 vs 32±13 ; t=3.06,t=3.24; t=2.36,t=2.82,all P<0.05].Higher levels of IFN-γand Th1 cytokine were found in BALF supernatant of C.sinensis and NAC intervention groups (t=2.34,t=2.32,both P<0.05).Conclusion C.sinensis may present immunomodulatory effects on Th1/Th2 imbalance via up-regulation of glutathione within macrophages in COPD models and supplementation of anti-oxidization capacity.
