目的 探讨成纤维细胞生长因子10(FGF10)在人类后肠及肛门直肠形态发生过程中的表达方式及可能的作用.方法 收集发育正常的人胚胎标本85例,通过矢状面连续切片免疫组化染色试验方法,观察FGF10在人类胚胎胎龄3~8周的泄殖腔以及肛门直肠时空性表达模式.结果 人类胚胎胎龄4~7周,泄殖腔分化过程中,FGF10阳性细胞主要集中在泄殖腔膜、顶端及背侧尿直肠隔.待第七周时,泄殖腔膜破裂后,肛门直肠及尿生殖窦与羊膜腔相通,FGF10阳性细胞在肛管上皮消失,但尿道上皮出现FGF10阳性细胞.结论 在人类肛门直肠发育过程中,FGF10在泄殖腔膜、背侧尿直肠隔表达阳性;当肛门直肠形成后,FGF10在肛门区域不再表达,提示FGF10在人类肛门直肠发育过程中可能发挥重要作用.
目的 探討成纖維細胞生長因子10(FGF10)在人類後腸及肛門直腸形態髮生過程中的錶達方式及可能的作用.方法 收集髮育正常的人胚胎標本85例,通過矢狀麵連續切片免疫組化染色試驗方法,觀察FGF10在人類胚胎胎齡3~8週的洩殖腔以及肛門直腸時空性錶達模式.結果 人類胚胎胎齡4~7週,洩殖腔分化過程中,FGF10暘性細胞主要集中在洩殖腔膜、頂耑及揹側尿直腸隔.待第七週時,洩殖腔膜破裂後,肛門直腸及尿生殖竇與羊膜腔相通,FGF10暘性細胞在肛管上皮消失,但尿道上皮齣現FGF10暘性細胞.結論 在人類肛門直腸髮育過程中,FGF10在洩殖腔膜、揹側尿直腸隔錶達暘性;噹肛門直腸形成後,FGF10在肛門區域不再錶達,提示FGF10在人類肛門直腸髮育過程中可能髮揮重要作用.
목적 탐토성섬유세포생장인자10(FGF10)재인류후장급항문직장형태발생과정중적표체방식급가능적작용.방법 수집발육정상적인배태표본85례,통과시상면련속절편면역조화염색시험방법,관찰FGF10재인류배태태령3~8주적설식강이급항문직장시공성표체모식.결과 인류배태태령4~7주,설식강분화과정중,FGF10양성세포주요집중재설식강막、정단급배측뇨직장격.대제칠주시,설식강막파렬후,항문직장급뇨생식두여양막강상통,FGF10양성세포재항관상피소실,단뇨도상피출현FGF10양성세포.결론 재인류항문직장발육과정중,FGF10재설식강막、배측뇨직장격표체양성;당항문직장형성후,FGF10재항문구역불재표체,제시FGF10재인류항문직장발육과정중가능발휘중요작용.
Objective To determine the spatiotemporal expression of the fibroblast growth factor 10 (FGF10) in human embryos during hindgut and anorectal development,and to elucidate its possible role during morphogenesis of the human hindgut and anorectum.Methods The embryos (n =85)were sectioned serially and sagittally,and the expression of FGF10 on the caudal midline from the 3rd 8th weeks of gestation was shown by immunohistochemical staining.Results From the 4th to 7th week of gestation,the FGF10-positive cells were mainly located on the epithelium of the apical urorectal septum,hindgut,and cloacal membrane.After the anorectum and the urogenital sinus (UGS) opened to the amniotic cavity during the 7th week,the FGF10-positive cells disappeared and remained negative up to the 8th week on the epithelium of the anal canal.Conclusions The expression of FGF10 is constantly active during human hindgut and anorectum development and disappears after the anus formed,suggesting that FGF10 plays an important role in human hindgut and anorectal morphogenesis.
