目的 体外观察尿道下裂患儿阴茎包皮内板组织成纤维细胞生长特点,探讨成纤维细胞生长发育异常与尿道下裂发生的关系.方法 组织块法原代培养尿道下裂患儿的阴茎包皮内板组织成纤维细胞以及细胞增殖试验(CCK8法)检测其增值情况和流式细胞术分析其细胞周期.结果 在细胞原代培养实验中近端型尿道下裂组的成纤维细胞移出组织块的时间(9.5±1.0)d明显长于中远端型组(4.8±0.8)d和对照组(4.0±0.6)d(P值均<0.05),同时细胞生长融合成束所需时间也较中远端型组和对照组明显延长(P值均<0.001);在细胞增殖实验及细胞周期分析中显示近端型组尿道下裂受检的成纤维细胞同样表现为细胞增殖明显低于中远端型组和对照组[(17.1±2.2)d比(10.5±1.4)d比(9.0±1.1)(P值均<0.001)],中远端型低于对照组(P<0.05).结论 尿道下裂患儿生殖器成纤维细胞生长和增殖能力下降,可能是尿道下裂,尤其是近端型尿道下裂发病的重要因素之一,提示成纤维细胞功能越差,尿道下裂临床表现的程度可能越严重.
目的 體外觀察尿道下裂患兒陰莖包皮內闆組織成纖維細胞生長特點,探討成纖維細胞生長髮育異常與尿道下裂髮生的關繫.方法 組織塊法原代培養尿道下裂患兒的陰莖包皮內闆組織成纖維細胞以及細胞增殖試驗(CCK8法)檢測其增值情況和流式細胞術分析其細胞週期.結果 在細胞原代培養實驗中近耑型尿道下裂組的成纖維細胞移齣組織塊的時間(9.5±1.0)d明顯長于中遠耑型組(4.8±0.8)d和對照組(4.0±0.6)d(P值均<0.05),同時細胞生長融閤成束所需時間也較中遠耑型組和對照組明顯延長(P值均<0.001);在細胞增殖實驗及細胞週期分析中顯示近耑型組尿道下裂受檢的成纖維細胞同樣錶現為細胞增殖明顯低于中遠耑型組和對照組[(17.1±2.2)d比(10.5±1.4)d比(9.0±1.1)(P值均<0.001)],中遠耑型低于對照組(P<0.05).結論 尿道下裂患兒生殖器成纖維細胞生長和增殖能力下降,可能是尿道下裂,尤其是近耑型尿道下裂髮病的重要因素之一,提示成纖維細胞功能越差,尿道下裂臨床錶現的程度可能越嚴重.
목적 체외관찰뇨도하렬환인음경포피내판조직성섬유세포생장특점,탐토성섬유세포생장발육이상여뇨도하렬발생적관계.방법 조직괴법원대배양뇨도하렬환인적음경포피내판조직성섬유세포이급세포증식시험(CCK8법)검측기증치정황화류식세포술분석기세포주기.결과 재세포원대배양실험중근단형뇨도하렬조적성섬유세포이출조직괴적시간(9.5±1.0)d명현장우중원단형조(4.8±0.8)d화대조조(4.0±0.6)d(P치균<0.05),동시세포생장융합성속소수시간야교중원단형조화대조조명현연장(P치균<0.001);재세포증식실험급세포주기분석중현시근단형조뇨도하렬수검적성섬유세포동양표현위세포증식명현저우중원단형조화대조조[(17.1±2.2)d비(10.5±1.4)d비(9.0±1.1)(P치균<0.001)],중원단형저우대조조(P<0.05).결론 뇨도하렬환인생식기성섬유세포생장화증식능력하강,가능시뇨도하렬,우기시근단형뇨도하렬발병적중요인소지일,제시성섬유세포공능월차,뇨도하렬림상표현적정도가능월엄중.
Objective Fibroblasts derived from inner prepuce tissue of hypospadias children and to discuss the relationship between unusual growth of fibroblasts and hypospadias.Methods Fibroblasts from inner plate of prepuce tissue of hypospadias children were primary cultured in-vitro.Their growth was examined using cell multiplication experiment(CCK8).Cc was analyzed by flow cytometry.Results We found that the fibroblasts the fibroblasts the fibroblasts group needed significantly more time to climb from the edge of the tissue than the distal and middle hypospadias group and the control group(P<0.05).They also needed more time to grow into beam(P<0.001).Cell growth experiment and the cell cycle analysis showed that fibroblasts of proximal hypospadias group had significantly lower multiplication capacity than the distal and middle group and the control group(P<0.001),while the growth rate of fibroblasts from distal and middle group was lower than the control group(P<0.05).Conclusions The reduction of growth and proliferation ability of fibroblasts in hypospadias children maybe one of the important factors of hypospadias,especially in proximal hypospadias.
