目的 以自建的人神经母细胞瘤转移瘤细胞(QDDQ-NM1)及原位瘤细胞(QDDQ-NM2)为研究对象探讨KAI1/CD82在不同转移潜能人神经母细胞瘤细胞系细胞中的表达情况.方法 采用RT-PCR检测两神经母细胞瘤细胞系细胞KAI1 mRNA的表达;免疫组化方法检测细胞CD82的表达.结果 RT-PCR显示QDDQ-NM1细胞系KAI1 mRNA的相对含量为0.0748±0.0063,QDDQ-NM2细胞系为0.4576±0.0226,两者比较差异有统计学意义(P<0.05),免疫组化法检测两细胞系细胞CD82蛋白表达的阳性率分别为30%和75%,两者的差异有统计学意义(P<0.05).结论 神经母细胞瘤细胞系的细胞高表达KAI1/CD82,与人神经母细胞瘤细胞系细胞的体外高转移潜能呈负相关.

Objective To study the expression of KAI1/CD82 in the human neuroblastoma with different metastatic potential.Methods Neuroblastoma NB metastatic tumor (QDDQ-NM) and primary tumor (QDDQ-NM2) cell lines were used in this study.The mRNA expression of KAI1 was determined by reverse transcription-polymerase chain reaction (RT-PCR).The protein level of CD82 was studied by immunocytochemical staining.Results The mRNA relative expression level of KAI1 in QDDQ-NM1 cell lines was 0.074 8 ± 0.006 3,which was significantly higher than that in QDDQ-NM2 cell lines (0.074 8 ± 0.006 3 vs 0.457 6 ± 0.022 6,P<0.05).The CD82 immunoreactive cells was 30% in QDDQ-NM1 cell lines and 75% in QDDQ-NM2 cell lines,the difference between the 2 groups was significant.Conclusions The expression of KAI1/CD82 is negatively correlated with the metastatic potential of human neuroblastoma.
