目的 检测骨形成蛋白4 (bone morphogenetie protein 4,BMP4)在先天性肾盂输尿管连接部梗阻(ureteropelvic junction obstruction,UPJO)所致肾积水肾盂输尿管连接部组织中的表达,探讨其在该病发生中的作用及机制.方法 收集先天性UPJO患儿手术中切除的肾盂输尿管连接部组织30例作为病例组,同期收集肾母细胞瘤未受肿瘤侵犯的正常肾盂输尿管连接部组织18例作为对照组,应用反转录-实时荧光定量PCR(RT qPCR)、免疫组化S-P法及Western blot检测BMP4的表达.结果 BMP4主要表达于输尿管的黏膜及肌肉层,在病例组中的表达明显低于对照组(病例组mRNA,黏膜层、肌层蛋白质相对表达量分别为:0.53±0.27、0.16±0.05、0.14±0.03;对照组的分别为:1.23±0.46、0.26±0.03、0.18±0.04),差异有统计学意义(P<0.05).结论 BMP4参与了肾脏输尿管的发育,其表达降低可能与UPJO的发生相关.
目的 檢測骨形成蛋白4 (bone morphogenetie protein 4,BMP4)在先天性腎盂輸尿管連接部梗阻(ureteropelvic junction obstruction,UPJO)所緻腎積水腎盂輸尿管連接部組織中的錶達,探討其在該病髮生中的作用及機製.方法 收集先天性UPJO患兒手術中切除的腎盂輸尿管連接部組織30例作為病例組,同期收集腎母細胞瘤未受腫瘤侵犯的正常腎盂輸尿管連接部組織18例作為對照組,應用反轉錄-實時熒光定量PCR(RT qPCR)、免疫組化S-P法及Western blot檢測BMP4的錶達.結果 BMP4主要錶達于輸尿管的黏膜及肌肉層,在病例組中的錶達明顯低于對照組(病例組mRNA,黏膜層、肌層蛋白質相對錶達量分彆為:0.53±0.27、0.16±0.05、0.14±0.03;對照組的分彆為:1.23±0.46、0.26±0.03、0.18±0.04),差異有統計學意義(P<0.05).結論 BMP4參與瞭腎髒輸尿管的髮育,其錶達降低可能與UPJO的髮生相關.

Objective To study the expression of BMP4 in the ureteropelvic junction of patients with congenital ureteropelvic junction obstruction (UPJO).Methods Samples of ureteropelvic junction were collected from 30 patients with congenital UPJO (patient group) and from 18 patients with Wilms' tumor (control group).The expression of BMP4 was detected using RT-qPCR,immunohisto chemistry (IHC) and western blot (WB).Results BMP4 was expressed in the mucosal layer and smooth muscle layer.However,the expression levels of BMP4 mRNA and protein were significantly lower in the patient group.The relative expressions of BMP4 mRNA and protein in the patient group were 0.53 ± 0.27,0.16 ± 0.05 (mucosa) and 0.14 ± 0.03 (smooth muscle) respectively,which were significantly lower than that of the control group (1.23 ± 0.46,0.26 ± 0.03 and 0.18 ± 0.04 respectively).Conclusions BMP4 participates in the process of nephrogenesis.Abnormal expression of BMP4 may play important roles in the pathogenesis of UPJO.
