目的 通过对钛合金表面磨砂、酸蚀、碱热的复合处理,观察其对成骨细胞生物学行为的影响. 方法 将商用钛合金表面进行磨砂、酸蚀及碱热处理,分为表面光滑组(对照组),表面磨砂酸蚀(sandblasted and acid-etched,SLA)组(SLA组)和SLA-碱热(alkli-heat,AH)组(SLA-AH组),SD大鼠成骨细胞分别与三组钛合金体外共同培养.用细胞计数、细胞增殖实验、扫描电镜观察钛合金表面的成骨细胞黏附能力、增值、形态变化,采用实时定量RT-PCR(real-time RT-PCR,qRT-PCR)比较三组钛合金表面成骨细胞ALP、骨钙素、Ⅰ型胶原及核心结合因子α1(core binding factor alpha 1,Cbfα1)mRNA合成含量. 结果 共同培养1,2,4h后,黏附能力实验结果显示,早期SLA-AH组黏附的成骨细胞数多于其他两组(F =5.635,28.621,124.037,P<0.05).细胞增殖实验显示,在共同培养第7天,与对照组比较,SLA组、SLA-AH组吸光度值较小(F=12.682,P<0.05).扫描电镜观察显示,SLA-AH组表面成骨细胞比其他两组更具有典型形态特征,伸展良好,更具有丰富的板状、丝状伪足.qRT-PCR结果显示,共培养第7天ALP及Cbfa1在SLA-AH组表达量较SLA组及对照组高,而在第14天较对照组低,骨钙素、Ⅰ型胶原表达量在第14天较SLA组及对照组高. 结论 经磨砂、酸蚀、碱热复合处理的钛合金表面能促进大鼠成骨细胞黏附、分化,但不能促进成骨细胞增殖.

Objective To process the titanium surface via sand-blasting,acid-etching and alkali-heating and investigate its effect on biological behavior of the osteoblasts.Methods The osteoblasts from SD rats in vitro were cultured in vitro on smooth titanium surface (control group),sandblasted and acid-etched (SLA) titanium surface (SLA group),and SLA plus alkali-heated (AH) titanium surface (SLA-AH group) respectively.Adhesion ability,proliferation and morphological changes of osteoblasts on titanium surface were observed respectively by cells counting,cell proliferation assay and scanning electron microscope.Levels of alkaline phosphatase (ALP),osteocalcin,type Ⅰ collagen,and core binding factor alpha 1 (Cbfαl) mRNA on titanium surface were compared among groups by real-time quantitative polymerase chain reaction (qRT-PCR).Results After 1,2,and 4 hours of co-culture,early-stage adhesion of osteoblasts in SLA-AH group was more than that in other two groups (F =5.635,28.621,124.037,P < 0.05).In adhesion assay,the decreased absorbance value was observed in SLA and SLA-AH groups at day 7 compared with control group (F =12.682,P < 0.05).Scanning electron microscopy showed oste oblasts in SLA-AH group presented better typical morphological characteristics,more stretch,and more lamellipodia and filopodia than other two groups.qRT-PCR results showed levels of ALP and Cbfa1 in SLA-AH group were higher than those in SLA group and control group at day 7,but lower than those in control group at day 14 and that levels of osteocalcin and collagen Ⅰ in SLA-AH group were higher than those in SLA and control groups at day 14.Conclusion SLA-AH-processed titanium surface enhances the attachment and differentiation of the rat osteoblasts,but fails to promote the proliferation of the osteoblasts.
