目的 初步探讨AIP基因突变与垂体瘤侵袭性的相关性.方法 根据Knosp分级标准筛选出侵袭性垂体瘤18例,非侵袭性垂体瘤16例.两组共34例垂体瘤标本分别行PCR和AIP基因6个外显子的DNA测序,根据测序比对结果,对两组样本中AIP基因的突变进行统计学分析.结果 18例侵袭性垂体瘤中有4例出现Exon4的突变,而16例非侵袭性垂体瘤中仅有1例出现突变,前者AIP基因突变率为22%,后者为6%.经卡方检验,两组样本中的突变率差异有统计学意义(P=0.001).突变位点是Exon 4序列中一处C突变为T的突变位点.结论 侵袭性垂体瘤中AIP基因的突变率要显著高于非侵袭性垂体瘸,表示AIP基因的突变与垂体瘤的侵袭性之间存在相关性;Exon 4序列中有一处C突变为T的突变位点,表示样品中位于Exon 4该位点的基因型为CT杂合型;AIP基因有可能成为诊断并治疗侵袭性垂体瘤的新靶点.

Objective To analyze the relationship between the mutations of AIP gene and the aggressive ability of pituitary adenomas.Method The clinical data of 18 invasive pituitary adenomas and 16 non - invasive pituitary adenomas classified by Knosp criteria were collected.PCR and DNA sequence of six exons in AIP of all samples were performed.Results Through comparing the sequence investigated with subjects in Genebank,it was found that there were 4 of 18 (22%) specimens mutated in aggressive pituitary adenomas while there was 1 of 16( 6% ) mutated in other samples.There was significantly difference between the two samples ( P =0.001 ).The mutation types were transferred from C to T.Conclusions There is higher mutation rate of AIP gene in aggressive pituitary tumors than that in non - invasive tumors.There is relationship between AlP mutations and aggression.In the mutation site,the base is transferred from C to T.It is suggested that the genotype is mixed of C and T.The relationship between AIP mutations and aggression need more samples to certify.AIP gene will be expected to become a new target point for early diagnosis and therapy of aggressive pituitary adenomas.
