目的 探讨肿瘤坏死因子凋亡相关诱导配体(TRAIL)与硝普钠(SNP)联合诱导U251细胞凋亡的协同作用.方法 将TRAIL和SNP单药或联合作用于U251胶质瘤细胞系.MTT比色法、流式细胞术检测细胞生长抑制率、凋亡率;Western blot法检测DR5、Caspase-3、Bcl-2、Survivin 蛋白表达.结果 TRAIL和SNP单药组对U251细胞活性均有抑制作用;联合作用组细胞凋亡率明显高于单药组和对照组(P<0.01),并存在协同作用;联合作用组较对照组DR5、Caspase-3蛋白表达明显上调,Survivin、Bcl-2蛋白表达明显下调(P<0.01).结论 TRAIL和SNP可显著抑制U251细胞增殖,具有协同细胞凋亡诱导作用.其机制与上调DR5、Caspase-3,下调Bcl-2、Survivin 蛋白表达有关.

Objective To investigate the synergistic apoptotic effect of TNF-Related Apoptosis,inducing Ligand (TRAIL) combined with sodium nitroprusside (SNP).Methods U251 glioma cell line was treated with TRAIL,SNP respectively and their combination.The inhibition rate of U251 cell line was detected with MTT assay,and the apoptotic rate was analyzed with flow cytometry.The protein expressions of DR5、Caspase-3、Bcl-2、survivin were detected with western blot.Results Inhibitory effect of U251 cell line was observed in both TRAIL and SNP groups,and the combination group represented significantly higher inhibition rate and apoptotic rate.Synergistic apoptosis inducing effect was observed in the combination group (P <0.01).Compared with control,the expression of Caspase-3 was higher,while that of Bcl-2 was lower in both groups,and the expression of DR5 was higher while that of survivin was lower in the SNP group (P <0.01).Conclusions TRAIL and SNP can inhibit the proliferation and induce apoptosis of U251 cell line.Their combination has a synergistic effect which may due to the up-regulation of DR5 and Caspase -3 and down-regulation of Bcl-2 and survivin.
