目的 探讨高迁移率族蛋白A2 (HMGA2)和基质金属蛋白酶(MMP-2)在星形细胞瘤中的表达与侵袭性、病理学分型的关系及HMGA2和MMP-2之间的相互关系.方法 应用免疫组化(S-P)方法对78例星形细胞瘤组织中的HMGA2和MMP-2进行检测.应用实时荧光定量聚合酶链反应(qPCR)检测HMGA2和MMP-2的mRNA表达.结果 78例星形细胞瘤中HMGA2和MMP-2的蛋白阳性率分别为92.30%和89.74%,HMGA2基因表达水平在胶质母细胞瘤(2.56倍,P< 0.01)和间变性星形细胞瘤(1.98倍,P<0.01)中显著高于星形细胞瘤,而在胶质母细胞瘤与间变性星形细胞瘤之间无显著区别(1.29倍,P=0.118).HMGA2与MMP-2之间的蛋白及mRNA表达水平存在相关关系(P<0.01).结论 HMGA2及MMP-2的增强表达与星形细胞瘤恶性程度增高及肿瘤侵袭性增强有密切关系,其表达可作为判断星形细胞瘤患者预后的指标.

Objective To probe into the expression of high mobility group A2 (HGMA2) and matrix metalloproteinase-2 (MMP-2) in low-grade and high-grade astrocyte tumors and their correlation with invasion,pathological classification and their reciprocal relationship.Methods The expression of HMGA2 and MMP-2 was examined by immunohistochemical staining method (S-P).The mRNA expression of HMGA2 and MMP-2 was examined with RT-PCR.Results The positive cell rates of HMGA2 and MMP-2 in 78 astrocyte tumors were 92.30% and 89.74%,respectively,and the degree of positive cells was in accordance with the malignancy of tumors.The QPCR results showed that the expression of HMGA2 protein was significantly higher in glioblastoma multiforme (2.56 fold,P < 0.01) and anaplastic astrocytoma(1.98 fold,P < 0.01) tumors than that in diffuse astrocytoma.There was correlation between HMGA2 and MMP-2.Conclusions The up-regulated expression level of HMGA2 and MMP-2 was closely related to the increased malignancy and invasion of tumors.The expression of protein and mRNA might act as an indicator the prognosis of patients with astrocyte tumors.
