目的 探讨微小RNA(miR)-21在甲状腺乳头状癌细胞中(TPC-1)的作用.方法 在TPC-1中转染miR-21,免疫组织化学检测转染后程序性细胞凋亡4(PDCD4)表达;用噻唑蓝(MTT)比色法测转染miR-21后TPC-1细胞增殖;流式细胞术检测转染后TPC-1细胞凋亡.结果 经免疫组织化学检测转染组中,细胞胞质呈棕褐色,对照组均未见染色,转染组PDCD4的表达明显高于对照组,MTT法检测转染组细胞活力增加(P<0.05).转染组的凋亡率为(3.13±0.34)%,空白组、空质粒组的凋亡率分别为(8.03±0.02)%、(9.51±0.54)%(P<0.05),转染组细胞凋亡率明显减少.结论 PDCD4是miR-21的靶基因之一,miR-21的高表达可促进甲状腺乳头状癌的发展.

목적 탐토미소RNA(miR)-21재갑상선유두상암세포중(TPC-1)적작용.방법 재TPC-1중전염miR-21,면역조직화학검측전염후정서성세포조망4(PDCD4)표체;용새서람(MTT)비색법측전염miR-21후TPC-1세포증식;류식세포술검측전염후TPC-1세포조망.결과 경면역조직화학검측전염조중,세포포질정종갈색,대조조균미견염색,전염조PDCD4적표체명현고우대조조,MTT법검측전염조세포활력증가(P<0.05).전염조적조망솔위(3.13±0.34)%,공백조、공질립조적조망솔분별위(8.03±0.02)%、(9.51±0.54)%(P<0.05),전염조세포조망솔명현감소.결론 PDCD4시miR-21적파기인지일,miR-21적고표체가촉진갑상선유두상암적발전.
Objective To explore the roles of microRNA (miR)-21 in the thyroid papillary carcinoma (TPC-1) cells.Methods Transit in the TPC-1 transfection of miR-21,Immunohistochemical detection of programmed cell death 4 (PDCD4) expression after transfection;transfected with miR-21 TPC-1 cell proliferation measured by tetrazolium (MTT) assay; flow cytometry detection of transfected TPC-1 cells withereddie.Results By transfection group were detected by immunohistochemistry,cytoplasmic brown color,the control group showed no staining,transfected group programmed cell death 4 (PDCD4) expression was significantly higher than the methyl thiazol MTT assay transfected cell viability was significantly be promoted (P < 0.05).Transfection group apoptosis rate was (3.13 ± 0.34) %,and a blank group,the apoptosis rate of the empty plasmid group (8.03 ± 0.02) % (9.51 ± 0.54) % (P < 0.05),apoptosis of the transfected cells is significantly reduced.Conclusion PDCD4 is a miR-21 target genes of high expression of miR-21 can promote the development of papillary thyroid carcinoma.
