目的 探讨O6-甲基鸟嘌呤-DNA-甲基转移酶(MGMT)、SFN基因启动子区甲基化异常与皮肤鳞癌发生的关系及临床意义.方法 采用甲基化特异性聚合酶链反应(MSP)和逆转录-聚合酶链反应(RT-PCR)法检测40例皮肤鳞癌和20例正常皮肤组织中MGMT、SFN两组基因甲基化状态及mRNA的表达.结果 鳞癌组织中的MGMT、SFN基因启动子附近甲基化频率分别为42.5%、35.0%,明显高于正常皮肤组织的10.0%和5.0%,差异有统计学意义(P<0.05),而鳞癌组织中MGMT和SFN的mRNA表达则低于正常皮肤组织(P<0.05).结论 皮肤鳞癌组织中MGMT、SFN基因启动子区附近高甲基化与该肿瘤的发生密切相关.

Objective To detect the correlation between the abnormal methylation status of O6-methylguanine-DNA methytransferase (MGMT) and SFN with progress of the skin squamous cell carcinoma and the clinical implication.Methods The methylation status of MGMT and SFN and the expression of their mRNA were detected by using the methylation specific polymerase chain reaction (MSP),and reverse transcription-PCR (RT-PCR) in 40 cases of skin squamous cell carcinoma and 20 normal skin tissues.Results The hypermethylation frequency of the MGMT and SFN in squamous cell carcinoma was 42.5% and 35.0% respectively,which was only 10.0% and 5.0% respectively in normal tissues (P < 0.05),while the expression of MGMT and SFN mRNA in the skin of squamous cell carcinoma was lower than in the normal tissues (P < 0.05).Conclusion The hypermathylation status of MGMT and SFN is related closely with the occurrence of the skin squamous cell carcinoma.
