目的 观察鱼藤素对胃癌细胞化疗敏感性的影响.方法 胃癌SGC-7901细胞以1 μmol/L鱼藤素处理24h后,噻唑蓝(MTT)比色法检测对其对氟尿嘧啶处理48 h的敏感性变化;经鱼藤素联合氟尿嘧啶(12.5 mg/L)处理后形态学观察进一步验证MTT结果;提取1μmol/L鱼藤素处理24 h后细胞总蛋白,Western blot法检测相关蛋白表达水平的变化.结果 鱼藤素预处理后可明显增强氟尿嘧啶对SGC-7901细胞的抑制作用,其IC50值降至7.86 mg/L(P<0.05);增强氟尿嘧啶对SGC-7901细胞增殖抑制和凋亡诱导作用;鱼藤素可明显降低蛋白激酶B(Akt)的磷酸化水平.结论 鱼藤素可通过下调Akt的活性,增强SGC-7901细胞对氟尿嘧啶的敏感性,与化疗药物联用可能有助于提高抗肿瘤治疗效果.

Objective To investigate the chemosensitivity of gastric carcinoma cells to deguelin.Methods Human gastric carcinoma cell line SGC-7901 and 5-fluorouracil (5-Fu) were used in present study.The chemosensitivity of cells was determined by the methyl thiazol tetrazolium (MTT) assay after exposure to 1 μmol/L deguelin for 24 h.The results of MTT were further confirmed by morphological analysis of cells treated with 1 mol/L deguelin combined with 5-Fu (12.5 mg/L).Total protein of cells was extracted after treatment with 1 μmol/L deguelin for 24 h,and the expression of proteins related was detected by using Western blotting.Results After pretreatment with deguelin,IC50 of 5-Fu in SGC-7901 cells was reduced to 7.86 mg/L (P < 0.05).Meanwhile,pretreatment with deguelin increased the proliferation inhibition and apoptotic induction of 5-Fu.The phosphorylated Akt was significantly downregulated after treatment with duguelin.Conclusion Deguelin enhanced the sensitivity of SGC-7901 cells to 5-Fu via inhibiting the activation of Akt,and it might be a potential sensitizer to improve the effects of chemotherapy.
