目的 观察巨噬细胞对胃癌细胞生物学行为的影响,以探讨肿瘤相关性巨噬细胞(TAMs)在胃癌侵袭转移中的作用.方法 将人单核细胞株THP-1体外诱导为巨噬细胞,并与胃癌细胞株AGS共培养模拟胃癌微环境,观察并分析其对胃癌细胞增殖力和侵袭力的影响.结果 胃癌细胞与巨噬细胞共培养后,增殖能力有所减弱,倍增时间为(38.42±2.68)h,而未经共培养者倍增时间为(24.36±2.91) h(P <0.001);而体外侵袭实验显示胃癌细胞共培养后及未共培养时侵袭细胞计数分别为308±15和168±4,侵袭力增强(P<0.01).巨噬细胞对胃癌细胞的作用与其分泌细胞因子相关.THP-1细胞激活并分化为巨噬细胞后,培养上清中白细胞介素(IL)-8及IL-1β表达量分别增加了40和20倍(P<0.01);肿瘤坏死因子(TNF)-α表达量增加了8倍(P>0.05).结论 巨噬细胞可增强胃癌细胞的侵袭力,该作用可能与其分泌的细胞因子IL-8及IL-1β和TNF-α增高有关.

Objective To observe the biological effects on human gastric cancer cell line AGS exerted by macrophages and discuss the role of tumor-associated macrophages (TAMs) on gastric cancer invasion and metastasis.Methods Human monocytic cell line THP-1 were differentiated to macrophages in vitro,then cocultured with AGS cells to mimic the micro-environment of gastric cancer.The effects on proliferation and invasiveness activities of gastric cancer cells initiated by macrophages were assessed.Results The AGS cells cocultured with macrophages were shown decreased proliferation,and the cell doubhng time was (38.42 ± 2.68) h,which was (24.36 ± 2.91) h in those non-cocuhured (P < 0.001).In vitro invasion assay indicated that the invasive cell count of AGS cells cocultured with macrophages and those noncocultured was 308 ± 15 and 168 ±4 separately (P <0.01).The effects of macrophages on gastric cancer cells were related to the cytokines.The THP-1derived macrophages secreted interleukin (IL)-1β 20 fold higher (P <0.01),IL-8 40 fold higher (P <0.01) and tumor necrosis factor (TNF)-α 8 fold higher (P >0.05) than those THP-1 cells did.Conclusion macrophages could enhance the invasiveness of gastric cancer cells,whith may ralated to their inceased secretion of IL-1β,IL-8 and TNF-α.
