目的 探讨微小RNA-200a (miR-200a)在胃癌中的差异性表达及其对上皮-间质转化(EMT)的调控机制.方法 分别采用原位杂交和免疫组织化学检测64例不同级别胃癌组织和10例正常胃黏膜组织miR-200a的表达和EMT相关蛋白锌指E-盒结合同源异形盒-1(ZEB1)/ZEB2、E-钙黏蛋白(E-cadherin)、N-钙黏蛋白(N-cadherin)和β-连环素(β-catenin)的表达.结果 与正常胃黏膜组织比较,胃癌中miR-200a明显下调(P<0.05),WHOⅢ/Ⅳ级胃癌组织中miR-200a的表达低于WHO Ⅰ/Ⅱ级(P<0.05).与正常胃黏膜组织比较,ZEB1/ZEB2、N-caderin和β-catenin在胃癌组织中表达明显上调,而E-cadherin表达明显下调.Speamen等级相关分析得出,miR-200a与ZEB1/ZEB2、N-cadherin和β-catenin呈负相关(P<0.05),miR-200a与E-cadherin呈正相关(P<0.05).结论 miR-200a抑制EMT的可能机制是miR-200a通过作用于其靶点ZEB1/ZEB2,促进E-cadherin的表达.

목적 탐토미소RNA-200a (miR-200a)재위암중적차이성표체급기대상피-간질전화(EMT)적조공궤제.방법 분별채용원위잡교화면역조직화학검측64례불동급별위암조직화10례정상위점막조직miR-200a적표체화EMT상관단백자지E-합결합동원이형합-1(ZEB1)/ZEB2、E-개점단백(E-cadherin)、N-개점단백(N-cadherin)화β-련배소(β-catenin)적표체.결과 여정상위점막조직비교,위암중miR-200a명현하조(P<0.05),WHOⅢ/Ⅳ급위암조직중miR-200a적표체저우WHO Ⅰ/Ⅱ급(P<0.05).여정상위점막조직비교,ZEB1/ZEB2、N-caderin화β-catenin재위암조직중표체명현상조,이E-cadherin표체명현하조.Speamen등급상관분석득출,miR-200a여ZEB1/ZEB2、N-cadherin화β-catenin정부상관(P<0.05),miR-200a여E-cadherin정정상관(P<0.05).결론 miR-200a억제EMT적가능궤제시miR-200a통과작용우기파점ZEB1/ZEB2,촉진E-cadherin적표체.
Objective To investagate the expression of microRNA-200a (miR-200a) and the mechanisms responsible for epithelial-mesenchymal transition (EMT) regulated by miR-200a in gastric carcinoma.Methods The levels of miR-200a and EMT-associated hallmarkers zinc-finger E-box binding homeobox 1 (ZEB1)1/ZEB2,E-cadherin,N-caderin and β-catenin in 64 samples of gastric carcinoma tissue and 10 samples of normal gastric mucosa tissue were detected with fluorescence in situ hybridization and Immuno-histochemistry.Results Compared to those in normal gastric mucosa tissue,the expression of miR-200a was significantly down-regulated (P < 0.05).The level of miR-200a in gastric carcinoma of WHOⅢ/IV were significant lower than those of WHO Ⅰ / Ⅱ (P <0.05).Compared to those in normal gastric mucosa tissue,the expression of ZEB1/ZEB2,N-caderin and β-catenin were elevated,but the E-cadherin expression was down-regulation in the gastric carcinoma.The Spearmen correlation showed significant inverse correlations existed between miR-200a expression and those of ZEB1/ZEB2,N-cadherin and β-catenin,and a significant positive correlation existed between miR-200a expression and E-cadherin expression.Conclusion miR-200a may inhibits EMT and promote the expression the E-cadherin by targeting E-cadherin repressor ZEB1/ZEB2.
