目的 观察分化抑制因子1(Id-1)、Id-3和基质金属蛋白酶9(MMP-9) mRNA、蛋白在脑转移瘤组织中的表达,探讨它们在脑转移瘤形成和发展过程中的作用及机制.方法 对39例脑转移瘤标本采用逆转录-聚合酶链反应(RT-PCR)和免疫组织化学法,观察Id-1、Id-3、MMP-9 mRNA及蛋白表达,并探讨它们表达之间的相关性.结果 Id-1、Id-3、MMP-9 mRNA在脑转移瘤中的相对表达量分别为0.75±0.07、0.83 ±0.06、0.72±0.09,明显高于正常脑组织(P<0.05);三者蛋白在脑转移瘤中的阳性表达率分别为92.3%、94.9%、79.5%,与正常脑组织比较表达明显增高(P<0.05).Id-1、Id-3与MMP-9 mRNA及蛋白表达均呈显著正相关(r=0.569,P<0.01;r=0.386,P<0.05).结论 Id-1、Id-3、MMP-9在脑转移瘤的形成与发展中发挥重要作用,且三者存在一定的协同作用.

목적 관찰분화억제인자1(Id-1)、Id-3화기질금속단백매9(MMP-9) mRNA、단백재뇌전이류조직중적표체,탐토타문재뇌전이류형성화발전과정중적작용급궤제.방법 대39례뇌전이류표본채용역전록-취합매련반응(RT-PCR)화면역조직화학법,관찰Id-1、Id-3、MMP-9 mRNA급단백표체,병탐토타문표체지간적상관성.결과 Id-1、Id-3、MMP-9 mRNA재뇌전이류중적상대표체량분별위0.75±0.07、0.83 ±0.06、0.72±0.09,명현고우정상뇌조직(P<0.05);삼자단백재뇌전이류중적양성표체솔분별위92.3%、94.9%、79.5%,여정상뇌조직비교표체명현증고(P<0.05).Id-1、Id-3여MMP-9 mRNA급단백표체균정현저정상관(r=0.569,P<0.01;r=0.386,P<0.05).결론 Id-1、Id-3、MMP-9재뇌전이류적형성여발전중발휘중요작용,차삼자존재일정적협동작용.
Objective To observe the expression of differentiation inhibitor 1 (Id-1),Id-3 and matrix metalloproteinase-9 (MMP-9) in cerebral metastatic tumors and to explore their roles and mechanism in the formation and development of cerebral metastases.Methods The expression of Id-1,Id-3 and MMP-9 mRNA and protein in cerebral metastases was detected by using reverse transcription-polymerase chain reaction (RT-PCR) and immunohistochemistry.The correlation of their expression was assayed statistically.Results The relative expression levels of Id-1,Id-3 and MMP-9 mRNA were 0.75 ± 0.07,0.83 ± 0.06 and 0.72 ± 0.09 respectively in brain metastases,obviously higher than those in human normal brain tissues (P <0.05).The positive expression rate of Id-l,Id-3 and MMP-9 protein was 92.3%,94.9% and 79.5% respectively in brain metastases,obviously higher than those in in human normal brain tissues (P < 0.05).The expression of Id-1,Id-3 mRNA and protein had a positive correlation with that of MMP-9 in metastases (r =0.569,P < 0.01 ; r =0.386,P < 0.05).Conclusion The over-expression of Id-1,Id-3 and MMP-9 played an important biological function in the formation of brain metastatic tumor,and there was a synergistic effect among them to some extent.
