目的 观察槲皮素对兔缺血再灌注心肌还原型烟酰胺腺嘌呤二核苷酸磷酸(NOX2)、一氧化氮合酶(NOS)基因和蛋白表达的影响.方法 建立成年家兔心肌缺血再灌注模型,结扎左缘支30 min,再灌注12h.实验分4组:空白对照组(control)、缺血再灌注组(I/R)、槲皮素组(Que)、槲皮素+缺血再灌注组(I/R+ Que).采用实时定量聚合酶链反应(Real-time PCR)和Western blot方法检测心肌NOX2、内皮型一氧化氮合酶(eNOS)、诱导型一氧化氮合酶(iNOS) mRNA和蛋白的表达.结果 I/R组NOX2 mRNA和蛋白表达分别为1.73±0.41、3.46 ±0.84,iNOS mRNA和蛋白表达分别为0.29±0.11、0.29 ±0.11,较对照组明显增高(P<0.01),eNOS变化不明显(P<0.01).给予槲皮素后,Que组NOX2 mRNA和蛋白表达分别为0.40±0.08、0.56 ±0.28,eNOS mRNA和蛋白表达分别为0.22±0.07、0.95±0.23,iNOS mRNA和蛋白表达分别为0.02±0.01、0.87±0.31;I/R+ Que组NOX2 mRNA和蛋白表达分别为1.03 ±0.31、2.05 ±0.49,eNOS mRNA和蛋白表达分别为0.32±0.09、1.12±0.36,iNOS mRNA和蛋白表达分别为0.16 ±0.05、3.17±1.29;较对照组水平明显降低(P<0.01).结论 心肌缺血再灌注后,NOX2、iNOS表达增加;槲皮素明显减少心肌缺血再灌注后NOX2、iNOS和eNOS的表达.

Objective Investigate the effects of quercetin on phagocytic nicotinamide adenine dinucleotide phosphate (NADPH) oxidase (NOX2),endothelial nitricoxide synthase (eNOS) and inducible nitricoxide synthase (iNOS) after myocardial ischemia-reperfusion injury (MIRI) in rabbit.Methods New Zealand rabbits were subjected to 30 minutes of myocardial ischemia followed by 12 hours of reperfusion.They then were randomly assigned to four experimental groups:control,L/R,quercetin (Que),I/R + Que.Gene and protein expression of NOX2,eNOS and iNOS were compared.Results mRNA and protein expression of NOX2 in I/R were 0.29 ± 0.11,0.29 ±0.11,mRNA and protein expression of iNOS in I/R were 0.29 ± 0.11,0.29 ± 0.11.Both in Real-time polymerase chain reaction (Real-time PCR) and Westem blotting studies,myocardial ischemia reperfusion-induced NOX2 and iNOS expression were enhanced (P <0.01),but eNOS mRNA and protein in I/R hearts were not significantly different from that in control (P<0.01).After administration of quercetin,mRNA and protein expression of NOX2 in Que were 0.40 ± 0.08,0.56 ± 0.28,mRNA and protein expression of eNOS in Que were 0.22 ± 0.07,0.95 ±0.23,mRNA and protein expression of iNOS in Que were 0.02 ±0.01,0.87 ±0.31 ; mRNA and protein expression of NOX2 in I/R + Que were 1.03 ±0.31,2.05 ±0.49,mRNA and protein expression of eNOS in I/R + Que were 0.32 ± 0.09,1.12 ± 0.36,mRNA and protein expression of iNOS in I/R + Que were 0.16 ±0.05,3.17 ± 1.29.Administration of quercetin reduced NOX2,eNOS and iNOS mRNA and protein expression both in control and I/R heart (P < 0.01).Conclusion Gene and protein expression of NOX2 and iNOS were increased after MIRI.Quercetin not only inhibited myocardial ischemia reperfusioninduced NOX2 and iNOS mRNA and protein expression but also eNOS mRNA and protein expression.
