目的 观察白细胞介素-17 (IL-17)在糖尿病大鼠肾组织中的表达,探讨其与肾小管-间质病变的关系.方法 90只大鼠随机分为对照组(Con组)、1型糖尿病组(T1DM组)及2型糖尿病组(T2DM组),以50 mg/kg链脲佐菌素(STZ)制备1型糖尿病模型,以高脂高糖饮食及25 mg/kgSTZ制备2型糖尿病模型.于2周、4周末采用酶联免疫吸附试验(ELISA)检测血、尿中IL-17水平;4周末采用免疫组织化学染色及Western blot检测肾组织中IL-17及转化生长因子-β1(TGF-β1)水平.结果 糖尿病大鼠2、4周末血、尿中IL-17升高;IL-17在Con组肾组织无表达,仅在糖尿病大鼠肾小管-间质中表达,T1DM组0.2357±0.0410,T2DM组0.1809±0.0090(P<0.05);TGF-β1肾组织表达,T1DM组0.1412±0.0610,T2DM组0.2809±0.0420 (P<0.05);肾组织IL-17与尿液IL-17水平呈正相关(r=0.715,P<0.05).结论 IL-17参与糖尿病肾小管-间质病变的发生;IL-17和TGF-β1在1型和2型糖尿病发展为糖尿病肾病中的作用机制存在差异.

목적 관찰백세포개소-17 (IL-17)재당뇨병대서신조직중적표체,탐토기여신소관-간질병변적관계.방법 90지대서수궤분위대조조(Con조)、1형당뇨병조(T1DM조)급2형당뇨병조(T2DM조),이50 mg/kg련뇨좌균소(STZ)제비1형당뇨병모형,이고지고당음식급25 mg/kgSTZ제비2형당뇨병모형.우2주、4주말채용매련면역흡부시험(ELISA)검측혈、뇨중IL-17수평;4주말채용면역조직화학염색급Western blot검측신조직중IL-17급전화생장인자-β1(TGF-β1)수평.결과 당뇨병대서2、4주말혈、뇨중IL-17승고;IL-17재Con조신조직무표체,부재당뇨병대서신소관-간질중표체,T1DM조0.2357±0.0410,T2DM조0.1809±0.0090(P<0.05);TGF-β1신조직표체,T1DM조0.1412±0.0610,T2DM조0.2809±0.0420 (P<0.05);신조직IL-17여뇨액IL-17수평정정상관(r=0.715,P<0.05).결론 IL-17삼여당뇨병신소관-간질병변적발생;IL-17화TGF-β1재1형화2형당뇨병발전위당뇨병신병중적작용궤제존재차이.
Objective To investigate the association between the expression of interleukin-17(IL-17) in renal tissues of rats with diabetic nephropathy and tubulointerstitial lesions.Methods Ninety rats were randomly divided into three groups:control group,type 1 diabetic mellitus (T1 DM) group,and type 2 diabetic mellitus (T2DM) group.The rats in T1DM group were given 50 mg/kg streptozoticin (STZ),and those in T2DM group fed on a diet enriched with sucrose,triglyceride and cholesterol,and injected with 25 mg/kg STZ.At 2nd and 4th week,serum and urine IL-17 levels were determined by using enzyme-linked immunosorbent assay (ELISA).The expression of IL-17 and transforming growth factor-β1 (TGF-β1) in the kidney was detected by using immunohistochemisty staining and Western blotting.Results Serum and urine IL-17 levels were obviously increase in diabetic rats.The expression of IL-17 was detectable in renal tubulointerstitium of diabetic rats.The expression of IL-17 in T2DM and T1DM groups was 0.1809 ±0.0090 and 0.2357 ±0.0410 respectively (P <0.05).The expression of TGF-β1 in T2DM and T1DM groups was 0.2809 ± 0.0420 and 0.1412 ± 0.0610 respectively (P < 0.05).The expression of IL-17 in renal tubulointerstitium of diabetic rats was positively correlated with urine IL-17 (r =0.715,P <0.05).Conclusion The increase of IL-17 expression may play an important role in diabetes mellitus-in-duced tubulointerstitial lesions.IL-17 and TGF-β1 might exert different effects on pathogenesis and progression of diabetic nephropathy from T1DM and T2DM.
