目的探讨骶骨脊索瘤组织中生存素(Survivin)和半胱氨酰天冬氨酸特异性蛋白酶-3(Caspase-3)的表达相关性及其临床意义.方法应用免疫组织化学(SP)法检测30例人骶骨脊索瘤组织和10例癌旁组织中Survivin和Caspase-3的表达.结果 Survivin在骶骨脊索瘤中的阳性表达率为70.0％,在瘤旁组织中不表达,两者差异有统计学意义(P＜0.05),Survivin的表达与患者肿瘤有无局部浸润、是否复发相关(P＜0.05)；Caspase-3在骶骨脊索瘤中的阳性表达率为33.3％,显著低于瘤旁组织的90.0％ (P ＜0.05),Caspase-3的表达与患者肿瘤有无局部浸润、是否复发相关(P＜0.05).Survivin与Caspase-3表达呈显著负相关(r=-0.463,P＜0.05).结论 Survivin在骶骨脊索瘤中高表达,可能通过抑制Caspase-3活性,从而阻止细胞凋亡,促进肿瘤细胞增殖与肿瘤的发生、发展.

Objective To investigate the expression and the clinical significance of Survivin and cysteinyl aspartate-specific protease (Caspase)-3 in human sacral chordoma tissues.Methods The expression of Survivin and Caspase-3 was detected in 30 cases of human sacral chordoma tissues and 10 normal notochord tissues by using streptavidin biotin peroxidase (SP) immunohistochemistry.Results The positive expression rate of Survivin in chordoma was 70.0％,while none of 10 cases in the normal notochord tissues expressed Survivin,with significant difference between two groups (P ＜0.05).The positive rate of Survivin was correlated significantly with tumor invasion of the surrounding muscle and recurrence (P ＜ 0.05).The positive expression rate of Caspase-3 was significantly lower in chordoma (33.3％) than that in normal notochord tissues (90.0％),with significant difference between two groups (P ＜ 0.05).The positive rate of Caspase-3 was correlated significantly with tumor invasion of the surrounding muscle and recurrence (P ＜ 0.05).The expression of Survivin and VEGF was correlated negatively with each other (r =-0.463,P ＜ 0.05).Conclusion The high expression of Survivin in sacral chordoma,possibly through inhibition of Caspase-3 activity,may prevent apoptosis and promote tumor cell proliferation,which may play a significant role in cancer development,progression and prognosis.
