目的 观察Wistar大鼠脊髓损伤后,脊髓组织中趋化因子受体4(CXCR4)表达变化,探讨大鼠脊髓损伤后CXCR4在修复脊髓损伤中的作用.方法 Wistar雌性大鼠80只,使用改良的Impactor modelⅡ法制作大鼠脊髓损伤模型,据取材时间分为8组(术后1、2、3、4、7、14、28 d与对照组,n=10).分别使用苏木素-伊红(HE)染色及免疫组织化学法染色观察脊髓组织中CXCR4表达变化.结果 大鼠脊髓损伤后,脊髓灰质中的神经元、胶质细胞、巨噬细胞和室管膜细胞均有CXCR4表达量的增多,神经元与巨噬细胞较神经胶质细胞更为显著.损伤中心的阳性细胞数量大于损伤头端与尾端的阳性细胞数量.脊髓灰质中CXCR4阳性细胞数量明显多于白质.大鼠脊髓损伤后24 h即有CXCR4表达增多(145.00±3.46)/mm2,第3天脊髓灰质中CXCR4阳性细胞达到高峰(181.50±6.73)/mm2 .结论 大鼠脊髓损伤后,脊髓组织中CXCR4表达发生变化,并有时间和空间的分布特点.
目的 觀察Wistar大鼠脊髓損傷後,脊髓組織中趨化因子受體4(CXCR4)錶達變化,探討大鼠脊髓損傷後CXCR4在脩複脊髓損傷中的作用.方法 Wistar雌性大鼠80隻,使用改良的Impactor modelⅡ法製作大鼠脊髓損傷模型,據取材時間分為8組(術後1、2、3、4、7、14、28 d與對照組,n=10).分彆使用囌木素-伊紅(HE)染色及免疫組織化學法染色觀察脊髓組織中CXCR4錶達變化.結果 大鼠脊髓損傷後,脊髓灰質中的神經元、膠質細胞、巨噬細胞和室管膜細胞均有CXCR4錶達量的增多,神經元與巨噬細胞較神經膠質細胞更為顯著.損傷中心的暘性細胞數量大于損傷頭耑與尾耑的暘性細胞數量.脊髓灰質中CXCR4暘性細胞數量明顯多于白質.大鼠脊髓損傷後24 h即有CXCR4錶達增多(145.00±3.46)/mm2,第3天脊髓灰質中CXCR4暘性細胞達到高峰(181.50±6.73)/mm2 .結論 大鼠脊髓損傷後,脊髓組織中CXCR4錶達髮生變化,併有時間和空間的分佈特點.
목적 관찰Wistar대서척수손상후,척수조직중추화인자수체4(CXCR4)표체변화,탐토대서척수손상후CXCR4재수복척수손상중적작용.방법 Wistar자성대서80지,사용개량적Impactor modelⅡ법제작대서척수손상모형,거취재시간분위8조(술후1、2、3、4、7、14、28 d여대조조,n=10).분별사용소목소-이홍(HE)염색급면역조직화학법염색관찰척수조직중CXCR4표체변화.결과 대서척수손상후,척수회질중적신경원、효질세포、거서세포화실관막세포균유CXCR4표체량적증다,신경원여거서세포교신경효질세포경위현저.손상중심적양성세포수량대우손상두단여미단적양성세포수량.척수회질중CXCR4양성세포수량명현다우백질.대서척수손상후24 h즉유CXCR4표체증다(145.00±3.46)/mm2,제3천척수회질중CXCR4양성세포체도고봉(181.50±6.73)/mm2 .결론 대서척수손상후,척수조직중CXCR4표체발생변화,병유시간화공간적분포특점.
Objective To investigate the temporal and spatial distributions and the role of CXC chemokine receptor 4 (CXCR4) in the process of recovery after spinal cord injury (SCI) in Wistar rats.Methods Adult female Wistar rats (n =80) were randomly divided into eight groups:the groups of 1,2,3,4,7,14,28 days after SCI and control group (n =10 eacb).Immunohistochemical technique and hematoxylin-eosin staining technique were used to examine the changes of CXCR4 temporal and spatial distributions in the spinal cord of Wistar rats.Results After SCI in rats,the expression of CXCR4 in neurons,glial cells,macrophages and ependymal cells in spinal cord gray matter was increased,especially in neurons and macrophages.The number of the CXCR4 positive cells near the injury site was significantly greater than that in the rostral and caudal regions,while the gray matter contained more CXCR4 positive cells than white matter.Twenty-four h after SCI in rats,the expression of CXCR4 was increased (145 ± 3.46)/mm2,and on the 3rd day,the CXCR4 expression in the spinal cord gray matter reached the peak [(181.5 ±6.73)/mm2].Conclusion CXCR4 was widely expressed and presented characteristics of temporal and spatial distributions after SPI in rats.
