目的 观察盐酸戊乙奎醚对大鼠内毒素性急性肺损伤晚期糖基化终末产物受体(RAGE)表达的变化,探讨其与肺组织核因子-κB(NF-κB)水平变化的关系及对肿瘤坏死因子-α(TNF-α)、白细胞介素-1β(IL-1β)的影响.方法 健康雄性SD大鼠30只,体质量200~220 g,按随机数字表法将大鼠随机分为3组:对照组(C组)、内毒素组(LPS组)和盐酸戊乙奎醚干预组(P)组,每组10只.LPS组经尾静脉注射LPS(5 mg/kg),P组在尾静脉注射LPS后腹腔注射盐酸戊乙奎醚(1 mg/kg),C组注射等容生理盐水.6h后处死大鼠,酶联免疫吸附试验(ELISA)法检测左肺支气管肺泡灌洗液(BALF)中TNF-α、IL-1β含量,右肺下叶行苏木素-伊红(HE)染色并进行病理学评分,Western blot法检测BALF中RAGE及肺组织胞核NF-κB p65的表达.结果 与C组比较,LPS组肺损伤评分(12.28±2.42)明显增高(P<0.01),BALF中TNF-α、IL-1β、RAGE及肺组织中NF-κB p65分别为(167.27±23.61) ng/L、(114.38±15.14) ng/L、0.74±0.15、0.69±0.12,均明显增高(P<0.01);与LPS组比较,P组肺损伤评分(8.93±1.86)明显降低(P<0.05),BALF中TNF-α、IL-1β、RAGE及肺组织中NF-κB p65分别为(118.64±19.43) ng/L、(98.16±12.55) ng/L、0.43 ±0.08、0.49±0.10,均明显降低(P<0.05).结论 盐酸戊乙奎醚可能通过RAGE-NF-κB通路降低了炎性因子TNF-α、IL-1β的水平,从而对大鼠内毒素性急性肺损伤具有保护作用.

목적 관찰염산무을규미대대서내독소성급성폐손상만기당기화종말산물수체(RAGE)표체적변화,탐토기여폐조직핵인자-κB(NF-κB)수평변화적관계급대종류배사인자-α(TNF-α)、백세포개소-1β(IL-1β)적영향.방법 건강웅성SD대서30지,체질량200~220 g,안수궤수자표법장대서수궤분위3조:대조조(C조)、내독소조(LPS조)화염산무을규미간예조(P)조,매조10지.LPS조경미정맥주사LPS(5 mg/kg),P조재미정맥주사LPS후복강주사염산무을규미(1 mg/kg),C조주사등용생리염수.6h후처사대서,매련면역흡부시험(ELISA)법검측좌폐지기관폐포관세액(BALF)중TNF-α、IL-1β함량,우폐하협행소목소-이홍(HE)염색병진행병이학평분,Western blot법검측BALF중RAGE급폐조직포핵NF-κB p65적표체.결과 여C조비교,LPS조폐손상평분(12.28±2.42)명현증고(P<0.01),BALF중TNF-α、IL-1β、RAGE급폐조직중NF-κB p65분별위(167.27±23.61) ng/L、(114.38±15.14) ng/L、0.74±0.15、0.69±0.12,균명현증고(P<0.01);여LPS조비교,P조폐손상평분(8.93±1.86)명현강저(P<0.05),BALF중TNF-α、IL-1β、RAGE급폐조직중NF-κB p65분별위(118.64±19.43) ng/L、(98.16±12.55) ng/L、0.43 ±0.08、0.49±0.10,균명현강저(P<0.05).결론 염산무을규미가능통과RAGE-NF-κB통로강저료염성인자TNF-α、IL-1β적수평,종이대대서내독소성급성폐손상구유보호작용.
Objective To investigate the effects of penequinine hydrochloride on receptor for advanced glycation end products (RAGE) expression in rats with lipopolysaccharide (LPS)-induced acute lung injury,its correlation with nuclear factor-kappaB (NF-κB) in the lung tissue,and the effect on tumor necrosis factor-α (TNF-α) and interleukin-1β (IL-1β) expression.Methods 30 male SD rats were randomly divided into three groups:control group,LPS group and penequinine hydrochloride group (n =10 each).The rats in the LPS group and penequinine hydrochloride group were injected intravenously with LPS (5 mg/kg) via tail vein to induce acute lung injury.The rats in penequinine hydrochloride group were injected intraperitoneally with penequinine hydrochloride (1 mg/kg body weight),and those in control group with the same volume of normal saline.All rats were sacrificed at 6 h later.The levels of TNF-α and IL-1 β in left bronchoalveolar lavage fluid (BALF) were determined by using enzyme linked immunosorbent assay (ELISA).The expression of RAGE in BALF and NF-κB p65 in the right lung was detected by using Western blotting.Results As compared with control group,the ALI scores (12.28 ±2.42),and the expression of TNF-α [(167.27 ± 23.61) ng/L],IL-1β [(114.38 ± 15.14) ng/L],RAGE (0.74 ± 0.15) and NF-κB p65 (0.69± 0.12) were significantly increased in LPS group (P < 0.01).As compared with LPS group,the ALI scores (8.93 ± 1.86),and the expression of TNF-α [(118.64 ± 19.43) ng/L],IL-1β [(98.16 ± 12.55) ng/L],RAGE (0.43 ±0.08) and NF-κB p65 (0.49 ±0.10) were significantly reduced in penequinine hydrochloride group (P < 0.05).Conclusion Penequinine hydrochloride has protective effects on LPS-induced acute lung injury in rats through inhibiting the expression of RAGE.
