目的 观察在胃癌中同源盒(HOX)基因家族表达,探讨两者的相关性.方法 采用Affymetrix U133 plus 2.0人类全基因组表达谱芯片分析在胃癌及其对应正常黏膜中HOX基因家族的差异表达,并行统计学分析.结果 HOXA基因家族中HOXA10、HOXA13在胃癌组织中表达明显升高,高表达率均为91.7% (11/12),其TvsN Signal Log Ratio值分别为0.4、5.4、4.2、6.1、7.1、5.2、3.4、1.9、1.6、3.0、3.5、3.3和1、4.8、0.3、6.9、4.9、3.5、4.2、4.3、5.4、4.1、2.2、2.8.同时HOX基因家族中还有多个基因表达上调,其中HOXA家族中HOXA1、HOXA4在胃癌表达升高,表达率均为66.7% (8/12);HOXB家族中HOXB7在胃癌中表达升高,表达率为75.0% (9/12);HOXC家族中HOXC10也表达升高,表达率为58.3% (7/12);而HOXC5和HOXC8则在肿瘤中呈现低表达,表达率分别为66.7% (8/12)和58.3% (7/12).结论 HOX家族中多个基因在胃癌中差异表达,提示该基因家族与胃癌发生发展密切相关.

Objective To screen the expression of homeobox (HOX) gene family in gastric cancers and explore the relationships between them.Methods Differential expression of HOX genes in gastric cancers and their corresponding normal mucosa was detected by using whole human genome microarray (Affymetrix GeneChip U133 Arrays plus2.0) and then statistically analyzed.Results HOXA10 and HOXA13 were more significantly highly expressed in the cancerous tissues than in their normal mucosa,and their expression rate was 91.7% (11/12).Their detailed TvsN Signal Ratio was 0.4,5.4,4.2,6.1,7.1,5.2,3.4,1.9,1.6,3.0,3.5,3.3 and 1.0,4.8,0.3,6.9,4.9,3.5,4.2,4.3,5.4,4.1,2.2,2.8 respectively.Meanwhile,there were several other differentially expressed genes in HOX gene family.HOXA1,HOXA4,HOXB7 and HOXC10 had a higher expression in tumors than in their normal tissues,and their expression rate was 66.7% (8/12),66.7% (8/12),75.0% (9/12),58.3% (7/12) respectively.However,HOXC5 and HOXC8 were down-regulated in tumors,and their expression rate was 66.7% (8/12) and 58.3% (7/12).Conclusion Several differentially expressed HOX genes were found in gastric cancers and might be closely involved in the process of the gastric carcinogenesis.
