目的 探讨键合表阿霉素纳米胶束(EPI-NPs)在大鼠移植性肝癌模型肝内的分布及药效.方法 制备具有靶向控释作用的EPI-NPs,Walker-256细胞直接注射法制作大鼠移植性肝癌模型,通过肝动脉灌注给药(EPI含量均为2 mg/kg),利用组织荧光和匀浆荧光技术对比分析EPI-NPs和表阿霉素(EPI)在肝内的分布特点,并作药效对比.结果 EPI-NPs组肿瘤组织的EPI荧光强度高于肝组织(P<0.05),而EPI组则表现为平均分布;EPI-NPs组在肿瘤及肝内的EPI荧光强度均高于EPI组(P<0.05).EPI-NPs组大鼠肝移植瘤的体积抑瘤率及质量抑瘤率分别为72.96%和68.19%,EPI组的体积抑瘤率及质量抑瘤率分别为39.16%和35.33%,两者差异均有统计学意义(P<0.05).EPI-NPs组生存率高于EPI组(P<0.05).结论 EPI-NPs具有良好的肿瘤组织靶向性和抑瘤效果.
目的 探討鍵閤錶阿黴素納米膠束(EPI-NPs)在大鼠移植性肝癌模型肝內的分佈及藥效.方法 製備具有靶嚮控釋作用的EPI-NPs,Walker-256細胞直接註射法製作大鼠移植性肝癌模型,通過肝動脈灌註給藥(EPI含量均為2 mg/kg),利用組織熒光和勻漿熒光技術對比分析EPI-NPs和錶阿黴素(EPI)在肝內的分佈特點,併作藥效對比.結果 EPI-NPs組腫瘤組織的EPI熒光彊度高于肝組織(P<0.05),而EPI組則錶現為平均分佈;EPI-NPs組在腫瘤及肝內的EPI熒光彊度均高于EPI組(P<0.05).EPI-NPs組大鼠肝移植瘤的體積抑瘤率及質量抑瘤率分彆為72.96%和68.19%,EPI組的體積抑瘤率及質量抑瘤率分彆為39.16%和35.33%,兩者差異均有統計學意義(P<0.05).EPI-NPs組生存率高于EPI組(P<0.05).結論 EPI-NPs具有良好的腫瘤組織靶嚮性和抑瘤效果.

Objective To study the distribution and anti-tumor effect of epirubicin nanoparticles (EPI-NPs) in transplanted hepatocarcinoma rat model.Methods EPI-NPs with targeting and controlled release quality were prepared.The transplanted hepatocarcinoma rat models were made by direct injection of cancerous ascites with Walker-256 cells,and the drug was delivered to the rat model through hepatic arterial infusion (EPI content was 2 mg/kg).The distribution of EPI-NPs in the hepatocarcinoma rat model was observed by tissue fluorescence analysis and homogenate fluorescence analysis.The anti-tumor effect of EPI-NPs was also studied.Results The intensity of drug fluorescence in tumor tissue was stronger in EPI-NPs group than that in normal tissue at different time points (P < 0.05).As to the EPI group,the fluorescence distributed equably in tumor and normal tissue.The intensity of drug fluorescence in tumor and intrahepatic tissue of EPI-NPs group was stronger than in EPIs group (P < 0.05).The volume anti-tumor ratio and the quality anti-tumor ratio in EPI-NPs group were 72.96% and 68.19% respectively,which were 39.16% and 35.33% in EPI group (P <0.05).The survival rate in EPI-NPs group was higher than that in EPI group.Conclusion EPI-NPs were proved to have good tumor targeting essence and favourable tumor suppression effect.
