目的 观察雌激素对骨髓细胞白细胞介素-6(IL-6)及骨髓源性破骨细胞的影响.方法 将90只雌性SD大鼠均分为3组:对照组(N组)、去卵巢组(OVX组)及雌激素组(OVX+ E2组).OVX组与OVX+ E2组大鼠行双侧卵巢切除术.术后OVX+ E2组按照0.02 ml/kg的剂量每周肌肉注射苯甲酸雌二醇1次.术后第1、3、5周取骨组织培养骨髓细胞,检测骨组织培养液及血清中IL-6含量,对破骨细胞进行计数.结果 (1)术后第1、3、5周OVX组骨组织培养液IL-6含量分别为(62.01±12.38)、(95.63±16.79)、(139.00±20.11) ng/L,OVX+E2组分别为(27.99±9.59)、(30.74±10.17)、(36.29±11.28) ng/L,两组含量均高于N组(P<0.05),且两组含量随时间逐渐增加(P<0.05);(2)各组各时间点血清中IL-6含量差异无统计学意义(P>0.05);(3)术后第1、3、5周OVX组破骨细胞数分别为(16.55±1.78)、(28.02±3.99)、(29.57±2.86)个/高倍视野,OVX+ E2组分别为(10.99±1.52)、(20.02±3.52)、(18.01±2.17)个/高倍视野,OVX+ E2组各时间点破骨细胞数均小于OVX组(P<0.05),但均大于N组(P<0.05).结论 雌激素对骨髓细胞IL-6具有一定抑制作用,但对血清中IL-6无明显影响;雌激素对骨髓源性破骨细胞形成具有一定抑制作用.

목적 관찰자격소대골수세포백세포개소-6(IL-6)급골수원성파골세포적영향.방법 장90지자성SD대서균분위3조:대조조(N조)、거란소조(OVX조)급자격소조(OVX+ E2조).OVX조여OVX+ E2조대서행쌍측란소절제술.술후OVX+ E2조안조0.02 ml/kg적제량매주기육주사분갑산자이순1차.술후제1、3、5주취골조직배양골수세포,검측골조직배양액급혈청중IL-6함량,대파골세포진행계수.결과 (1)술후제1、3、5주OVX조골조직배양액IL-6함량분별위(62.01±12.38)、(95.63±16.79)、(139.00±20.11) ng/L,OVX+E2조분별위(27.99±9.59)、(30.74±10.17)、(36.29±11.28) ng/L,량조함량균고우N조(P<0.05),차량조함량수시간축점증가(P<0.05);(2)각조각시간점혈청중IL-6함량차이무통계학의의(P>0.05);(3)술후제1、3、5주OVX조파골세포수분별위(16.55±1.78)、(28.02±3.99)、(29.57±2.86)개/고배시야,OVX+ E2조분별위(10.99±1.52)、(20.02±3.52)、(18.01±2.17)개/고배시야,OVX+ E2조각시간점파골세포수균소우OVX조(P<0.05),단균대우N조(P<0.05).결론 자격소대골수세포IL-6구유일정억제작용,단대혈청중IL-6무명현영향;자격소대골수원성파골세포형성구유일정억제작용.
Objective To investigate the effect of estrogen on interleukin 6 (IL-6) of the bone marrow cells and bone marrow-derived osteoclast formation.Methods Ninety 4-month-old female SD rats were randomly divided into three groups:sham operation group (N group),ovariectomized group (OVX group) and estrogen group (OVX + E2 group).Bone tissues and bone marrow cells were cultured of each group after treating 1,3,5 weeks.The levels of IL-6 in serum and the bone tissue culture fluid were tested,and bone-derived osteoclasts were numbered.Results (1) IL-6 levels of the OVX group were respectively (62.01 ± 12.38),(95.63 ± 16.79),(139.00 ± 20.11) after 1,3,5 weeks,and those were respectively (27.99 ± 9.59),(30.74 ± 10.17),(36.29 ± 11.28) ng/L of the OVX + E2 group,which were significantly higher than N group (P < 0.05),and the OVX and OVX + E2 group of bone tissue culture fluid IL-6 levels with treatment time increases (P < 0.05).(2) The serum IL-6 levels of all groups did not change significantly,and the difference was not statistically significant (P > 0.05).(3) After 1,3,5 weeks' treatment,the OVX group were respectively (16.55 ± 1.78),(28.02 ± 3.99),(29.57 ± 2.86) cells/HP,OVX + E2 group were respectively (10.99±1.52),(20.02±3.52),(18.01 ± 2.17) cells/HP,OVX + E2 treatment group at week 1,3,5 number of osteoclasts and bone resorption were significantly smaller than the number of OVX group (P < 0.05),but were more than N group.Conclusion The effect of estrogen on bone tissue IL-6 has a certain partial inhibition,but the serum levels of IL-6 had no significant effect; estrogen decreased the number of bone-derived osteoclast.
