目的 观察人胆囊癌组织中恶性脑瘤缺失1(DMBT1)基因的表达.方法 应用半定量逆转录-聚合酶链反应(RT-PCR)方法检测12例人中晚期胆囊癌及其癌旁正常组织中DMBT1mRNA的相对表达量,并采用Western blot检测上述组织中DMBT1蛋白表达.结果 91.7%(11/12)的胆囊癌组织中DMBT1 mRNA和蛋白表达下调或缺失,而癌旁组织中83.3%(10/12)该基因表达上调和蛋白表达增加,两者差异有统计学意义(P<0.O5).结论 胆囊癌组织中DMBT1表达处于下调或缺失状态,可能在胆囊癌的发生、发展过程中发挥重要作用.
目的 觀察人膽囊癌組織中噁性腦瘤缺失1(DMBT1)基因的錶達.方法 應用半定量逆轉錄-聚閤酶鏈反應(RT-PCR)方法檢測12例人中晚期膽囊癌及其癌徬正常組織中DMBT1mRNA的相對錶達量,併採用Western blot檢測上述組織中DMBT1蛋白錶達.結果 91.7%(11/12)的膽囊癌組織中DMBT1 mRNA和蛋白錶達下調或缺失,而癌徬組織中83.3%(10/12)該基因錶達上調和蛋白錶達增加,兩者差異有統計學意義(P<0.O5).結論 膽囊癌組織中DMBT1錶達處于下調或缺失狀態,可能在膽囊癌的髮生、髮展過程中髮揮重要作用.
목적 관찰인담낭암조직중악성뇌류결실1(DMBT1)기인적표체.방법 응용반정량역전록-취합매련반응(RT-PCR)방법검측12례인중만기담낭암급기암방정상조직중DMBT1mRNA적상대표체량,병채용Western blot검측상술조직중DMBT1단백표체.결과 91.7%(11/12)적담낭암조직중DMBT1 mRNA화단백표체하조혹결실,이암방조직중83.3%(10/12)해기인표체상조화단백표체증가,량자차이유통계학의의(P<0.O5).결론 담낭암조직중DMBT1표체처우하조혹결실상태,가능재담낭암적발생、발전과정중발휘중요작용.
Objective To observe the expression of deleted in malignant brain tumors 1 (DMBT1) gene in human gallbladder carcinoma.Methods The samples were obtained from 12 cases with gallbladder carcinoma specimens.Semi-quantitative reverse transcriptase-polymerase chain reaction (RT-PCR) strategies and Western blotting were used for analyzing the DMBT1 mRNA and protein expression in 12 samples and adjacent noncanerous tissues.Results DMBT1 mRNA and protein expression were not detected in 91.7% of human gallbladder carcinoma and in of 16.7% adjacent noncanerous tissues (P <0.05).Conclusion DMBT1 activation might be in human gallbladder carcinoma.DMBT1 gene may play some roles in the pathogenesis of human gallbladder carcinoma.
