目的 观察过表达磷脂酰肌醇蛋白聚糖-5(GPC5)对肺鳞癌细胞株SK-MES-1增殖、迁移和侵袭能力的影响.方法 构建慢病毒载体过表达GPC5-SK-MES-1和阴性对照NC-SK-MES-1稳转细胞株.通过细胞划痕和Transwell小室实验观察过表达GPC5对肺鳞癌细胞株SK-MES-1迁移和侵袭能力的影响.细胞计数试剂盒(CCK-8)法检测过表达GPC5对细胞增殖能力的影响,流式细胞仪检测其细胞周期的变化.结果 与阴性对照组NC-SK-MES-1细胞比较,过表达GPC5的肺鳞癌细胞的GPC5基因表达量(79.113±9.246)增加了86倍.其细胞迁移间距(729.89±46.15)较对照组的(438.33±19.77)显著增加;细胞穿膜数[(33±3)个]较对照组[(128±3)个]显著减少;细胞相对增殖活性较对照组显著降低;细胞周期检测显示S期细胞数比率[(50.32±0.97)%]较对照组[(32.59±1.54)%]显著增加,差异均有统计学意义(P<0.05).结论 上调肺鳞癌细胞GPC5的表达水平能够有效抑制肺鳞癌细胞的增殖、迁移和侵袭能力.

Objective To investigate the effects of the up-regulated expression of glypican-5 (GPC5) on the proliferation,migration and invasion of lung squamous carcinoma cell line SK-MES-1.Methods Lentivirus-induced GPC5 over-expression was used for in vitro functional analyses in lung squamous carcinoma cell line SK-MES-1,including migration and invasive assay,proliferation assay as well as cell cycle analysis.Results Compared to the control,the expression of GPC5 was increased by 86 times in GPC5-SK-MES-1,whose length of cell migration spacing was longer [(729.89 ±46.15) vs.(438.33 ±19.77)] and number of penetrating the membrane was significantly reduced [(33 ± 3) vs.(128 ± 3)].The relative cell proliferation activity was significantly decreased,and the cell cycle analysis showed that the ratio of S-phase cells was markedly increased in GPC5-SK-MES-1 cells as compared with controls [(50.32 ± 0.97) % vs.(32.59 ± 1.54) %] (P < 0.05).Conclusion Up-regulation of GPC5 can inhibit the proliferation,migration and invasion of lung squamous carcinoma cells.
