目的 探讨水溶性量子点硒化镉/硫化锌(CdSe/ZnS)对泌尿系肿瘤活细胞的毒性效应,观察其对泌尿系肿瘤细胞生长的影响.方法 将不同浓度的量子点(5、10、20、40、80 nmol/L)分别作用于膀胱癌EJ细胞和前列腺癌PC-3细胞,通过细胞计数试剂盒(CCK-8)检测法来评价其对肿瘤细胞活性的影响.结果 CCK-8检测发现,两种活体细胞和量子点共同孵育24 h后,其存活率随着量子点浓度的升高均显示下降趋势,在量子点通常使用浓度(5、10、20 nmol/L)下,两种细胞的存活率分别为101.0%、99.2%、97.9%(EJ细胞)和100.1%、101.0%、99.0%(PC-3细胞),与对照组比较差异无统计学意义(P>0.05),而在80 nmol/L高浓度时,两者存活率和对照组比较差异有统计学意义(P<0.001),但是两种细胞的平均存活率也高达92.6%(EJ细胞)和95.8%(PC-3细胞).结论 高浓度的量子点对膀胱癌EJ和前列腺癌PC-3细胞的活性有一定程度的影响,但是通常使用浓度的量子点对它们的存活率几乎没有影响.

Objective To detect the cytotoxic effect of quantum dots(CdSe/ZnS QDs) on the urinary tract tumor cells,Study the influence of CdSe/ZnS quantum dots on growth of the urinary tract tumor cells.Methods The cytotoxic effect of QDs in different concerntrations (5,10,20,40,80 nmol/L) was detected on bladder cancer cell line EJ and prostate cancer cell line PC-3 by cell counting kit-8 (CCK-8),assessing the activities of tumor cells.Results Two kinds of live cells incubated with QDs for 24 hours,using CCK-8 test,performed that the survival rate of cells was decreased with the increasing of QDs concentrations.However,in the common concentration (5,10 and 20 nmol/L),the survival rate of the two kinds of cells was 101%,99.2%,97.9% (EJ cell line)and 100.1%,101.0%,99.0% (PC-3 cell line) respectively.Moreover,no statistical difference in survival rate was observed between experimental and control groups (P > 0.05) ; in the high concentration (80 nmol/L),there were obvious difference between experimental and control groups (P < 0.01),however,the survival rates of bladder cancer cell line EJ and prostate cancer cell line PC-3 could still remained 92.6% and 95.8%,respectively.Conclusion The quantum dots in high concentrations have a certain degree of influence on the activity of bladder cancer cell line EJ and prostate cancer cell line PC-3,however,for the conventional use,the concentration of quantum dots have little impact on their survival.
