目的 检测SD大鼠膀胱癌组织的环氧合酶-2(COX-2)、毛细血管密度(MVD)、癌细胞凋亡水平,探讨其与病理分级的相关性.方法 采用N-甲基-N-亚硝基脲(MNU)构建模型,分为空白组、MNU组、华蟾素(Cino)组、阿霉素(ADM)组和卡介苗(BCG)组,每组10只.灌注结束1周后取其膀胱进行苏木素-伊红(HE)染色并检测相关指标.结果 免疫组织化学结果表明,低级别组中,各组COX-2的表达均无差异;Cino组在MVD表达和细胞凋亡两方面与ADM组相似,但均高于BCG组.在高级别组中,Cino组的COX-2和MVD表达均低于其余各组;而细胞凋亡则相反.结论 华蟾素能有效拮抗SD大鼠膀胱癌,且对高级别表达的膀胱癌具有抑制作用.

목적 검측SD대서방광암조직적배양합매-2(COX-2)、모세혈관밀도(MVD)、암세포조망수평,탐토기여병리분급적상관성.방법 채용N-갑기-N-아초기뇨(MNU)구건모형,분위공백조、MNU조、화섬소(Cino)조、아매소(ADM)조화잡개묘(BCG)조,매조10지.관주결속1주후취기방광진행소목소-이홍(HE)염색병검측상관지표.결과 면역조직화학결과표명,저급별조중,각조COX-2적표체균무차이;Cino조재MVD표체화세포조망량방면여ADM조상사,단균고우BCG조.재고급별조중,Cino조적COX-2화MVD표체균저우기여각조;이세포조망칙상반.결론 화섬소능유효길항SD대서방광암,차대고급별표체적방광암구유억제작용.
Objective To detect the expression of cyclooxygenase-2 (COX-2),microvascular density (MVD),and apoptosis of bladder cancer cells in the SD rats and explore their relationship with pathologic grade.Methods Fifty SD rats were divided randomly into 5 groups,including blank group,N-methyl-N-nitrosourea (MNU) group,cinobufagin (Cino) group,adriamycin (ADM) group and bacillus calmette-guerin (BCG) group.After perfusion for one week,bladder tissues were taken out for hematoxylin and eosin (HE) staining and detection of related indicators.Results Immunohistochemical results showed that COX-2 expression,in lower-grade groups,had no significant difference among different groups.MVD expression and apoptosis of cancer cells in Cino group and ADM group showed no significant difference,but higher than in BCG group.In the higher-grade group,COX-2 and MVD expression in Cino group (2.500 ± 0.535 and 11.750 ± 1.581 respectively) was lower than the rest groups,while apoptosis rate (18.500 ± 0.535) was higher than other groups.Conclusion Cinobufagin can effectively antagonize bladder cancer and suppress the growth of bladder cancer with high grade in SD rats.
