目的 观察糖尿病性膀胱病(DCP)豚鼠膀胱Cajal间质细胞(Icc)数量、形态和结构的改变,探讨ICC在DCP发病机制中的作用.方法 50只雌性豚鼠,随机选择42只作为实验组,8只作为对照组(C组).以单次腹腔注射链脲佐菌素法诱导实验组糖尿病豚鼠模型.尿动力学检查确定膀胱功能失代偿豚鼠(A组,即DCP组)及代偿豚鼠(B组,即代偿组).免疫组织化学荧光染色、激光共聚焦显微镜、透射电镜技术检测膀胱ICC形态、数量与结构变化.结果 A组膀胱ICC面积与膀胱组织面积百分比[(2.18±0.04)%]较C组[(2.81±0.04)%]及B组[(2.80±0.05)%]显著降低(P<0.01).DCP组膀胱ICC数量减少,细胞形态及超微结构发生明显改变.结论 糖尿病膀胱病豚鼠膀胱ICC数量减少、形态结构改变,提示ICC可能在DCP的发病过程中起重要作用.

Objective To investigate the bladder interstitial cells of Cajal (ICC) function in the pathogenesis of the diabetic cystopathy.Methods Fifty short hair Britain female guinea pigs were divided into two groups randomly,42 as the experiment group and the other 8 as the control group (group C).The experiment group was intraperitoneally injected with streptozotocin (STZ) to establish diabetic models.Diabetic guinea pigs were further divided into the decompensated bladder function group (namely DCP,group A) and compensated group (group B) by urodynamic study.Immunofluorescence staining,confocal laser microscope and transmission electron microscopy (TEM) technology were used to detect the morphology and ultrastructure changes of the ICC.Results The percentage of the ICC in group A was significantly lower than that of the group B and group C [(2.18 ± 0.04) % vs.(2.80 ± 0.05,2.81 ± 0.04) % respectively,P <0.01].Under the electron microscopic observation,the mitochondria in the ICC were reduced in the DCP group.The substrate also became less and shallow,and crest got shorter or even disappeared.Conclusion ICC may play an important role in the development of the DCP.
