目的 观察梯度神经生长因子(NGF)对背根神经节(DRG)轴突生长的引导及促进作用.方法 制备聚左旋丙交酯(PLLA)膜,再使NGF梯度负载于该膜.取孕14 d胎鼠DRG,随机分为两组,A组(n=10)种植于负载有均匀NGF的膜上;B组(n=10)种植于负载有梯度NGF的膜上.培养5d后,测量轴突长度.结果 梯度NGF组的DRG轴突长度为(1530±30).μm,轴突转弯系数(TR)为0.49 ±0.10;而均匀NGF组的DRG轴突长度为(1210±20)μm,TR为0.10±0.08.两组间的轴突长度与TR比较差异有统计学意义(P<0.05).结论 NGF梯度信号对背根神经节轴突生长具有引导及促进作用.

Objective To investigate the effects of gradient nerve growth factor (NGF) on guiding and promoting the growth of dorsal root ganglia (DRG) neurite.Methods The poly L-lactic acid (PLLA) membranes were prepared,and then coated by gradient NGF.Twenty DRGs from El4 rats were randomly divided into two groups.The DRGs in group A (n =10) were placed on the gradient NGF immobilized membranes,and those in group B (n =10) were placed on the uniform NGF immobilized membranes.After 5 days of culture,the effects of gradient NGF signal on the growth of neurites of DRGs were evaluated by using immunofluorescence staining.Results The length of DRG neurite in gradient NGF group was 1530 ±30 μm,and that in uniform NGF group was 1210 ±20 μm.The neurite turning ratio was 0.49 ± 0.10 in gradient NGF group,and that was 0.10 ± 0.08 in uniform NGF group.Conclusion The NGF gradient signal could promote and guide the growth of DRG neurite.
