目的 构建人肿瘤裸鼠移植瘤模型,探讨多亮氨酸重复区免疫球蛋白样蛋白3(LRIG3)表达沉默后对移植瘤生长以及表皮生长因子受体(EGFR)表达的影响.方法 将Hela229细胞接种于裸鼠背部皮下,成瘤后将裸鼠分为3组,分别在瘤体内注射生理盐水、LRIG3反义寡核苷酸(ASODN)、LRIG3错义寡核苷酸(MSODN),检测各组移植瘤生长以及LRIG3和EGFR表达.结果 ASOND组较其他两组瘤体生长加快、体积增大,苏木素-伊红(HE)染色可见核仁染色较深,核分裂相增多,LRIG3蛋白表达结果:空白对照组为0.2302±0.0108、MSODN组为0.2203±0.0183,ASODN组为0.1532±0.0132,两两比较,ASODN组肿瘤组织中LRIG3蛋白表达明显降低,差异有统计学意义(P<0.05).EGFR蛋白表达结果:空白对照组为0.9212±0.0796、MSODN组为0.9904±0.0830、ASODN组为1.2730±1.1116,两两比较,ASODN组肿瘤组织中EGFR蛋白表达明显增加,差异有统计学意义(P<0.05).LRIG3表达降低,EGFR表达升高.结论 LRIG3基因表达抑制引起人肿瘤裸鼠移植瘤生长加快,瘤体内EGFR表达升高.

Objective Leucine-rich repeats and Immunoglobulin-like domains (LRIGs) gene family is a group of recently discovered tumor-associated genes,which acts in a negatingve feedback loop of epidermal growth factor receptor (EGFR) and inhibits the deterioration and proliferation of cancer.The present study investigated the effect of LRIG3 silencing on tumor growth and EGFR expression of the human cervical carcinoma xenografts in nude mice.Methods The Hela229 cells were inoculated subcutaneously into nude mice.The mice were divided into three groups after intratumoral injection of saline,LRIG3 antisense oligodeoxynucleotide (ASODN),and LRIG3 MSODN,respectively.Results Compared to MSODN and saline groups,the tumors in ASOND group grew faster and showed larger size,and demonstrated more deeply stained nucleolus by HE staining.LRIG3 protein expression in the tumors of saline,MSODN and ASODN group were 0.2302 ± 0.0108,0.2203 ± 0.0183 and 0.1532 ± 0.0132,respectively,the expression was significantly down-regulated in ASODN group compared to the other two groups (P < 0.05).EGFR protein expression in the tumors in ASODN group was 1.2730 ± 1.1116,and significantly elevated compared to that in saline and MSODN group,which was 0.9212 ±0.0796 and 0.9904 ±0.0830,respectively (P < 0.05).Conclusion Inhibition of LRIG3 gene expression can accelerate human cervical carcinoma xenograft growth and elevate EGFR expression in xenograft tumors.
