目的 观察不同浓度的唑来膦酸/硫酸钙人工骨复合物体外释放规律.方法 分别制备不同浓度梯度唑来膦酸/硫酸钙人工骨复合物,用高效液相色谱法(HPLC)研究药物体外释放规律.结果 B~E组0至24 h有明显突释相,累计释放度均值分别达19.11%、23.45%、35.93%、26.88%,48 h以后出现缓释效应,C、D、E组3~21 d溶出率曲线出现波动,D、E组波动明显.随着唑来膦酸载药量的增加,硫酸钙结构逐渐不稳定.结论 载药量不超过硫酸钙人工骨最大载药量时,唑来膦酸在硫酸钙人工骨释放体系中能够持续、稳定的释放.
目的 觀察不同濃度的唑來膦痠/硫痠鈣人工骨複閤物體外釋放規律.方法 分彆製備不同濃度梯度唑來膦痠/硫痠鈣人工骨複閤物,用高效液相色譜法(HPLC)研究藥物體外釋放規律.結果 B~E組0至24 h有明顯突釋相,纍計釋放度均值分彆達19.11%、23.45%、35.93%、26.88%,48 h以後齣現緩釋效應,C、D、E組3~21 d溶齣率麯線齣現波動,D、E組波動明顯.隨著唑來膦痠載藥量的增加,硫痠鈣結構逐漸不穩定.結論 載藥量不超過硫痠鈣人工骨最大載藥量時,唑來膦痠在硫痠鈣人工骨釋放體繫中能夠持續、穩定的釋放.
목적 관찰불동농도적서래련산/류산개인공골복합물체외석방규률.방법 분별제비불동농도제도서래련산/류산개인공골복합물,용고효액상색보법(HPLC)연구약물체외석방규률.결과 B~E조0지24 h유명현돌석상,루계석방도균치분별체19.11%、23.45%、35.93%、26.88%,48 h이후출현완석효응,C、D、E조3~21 d용출솔곡선출현파동,D、E조파동명현.수착서래련산재약량적증가,류산개결구축점불은정.결론 재약량불초과류산개인공골최대재약량시,서래련산재류산개인공골석방체계중능구지속、은정적석방.
Objective To observe a drug delivery system based on calcium sulfate with the different concentrations of zoledronic acid in vitro.Methods Five concentrations of zoledronic acid were mixed with calcium sulfate and placed in distilled water.The concentration in the water was measured daily for 21 days,then the high performance liquid chromatography (HPLC) was used to observe the model of zoledronic release at different time points.Results The release of zoledronic acid was greatest during the first 24 h for all concentrations and decreased rapidly during next 24 h.The accumulated release rate of B,C,D and E was 19.11%,23.45%,35.93% and 26.88% respectively.The dissolution curve of the over drug-loaded groups waved seriously after 48 h.The structure of calcium sulfate was unstable with the increase of loading durgs.Conclusion Calcium sulfate is a good slow-release carrier of zoledronic within its range of maximum drug load.Zoledronic can release from calcium sulfate slowly and steay.
