目的 检测上皮钙黏蛋白(E-cadherin)和锌指转录抑制因子Snail在肝细胞癌(HCC)组织中的表达,探讨其在HCC发生发展中的意义.方法 采用实时定量聚合酶链反应(Real-timePCR)法检测36对HCC及癌旁组织中的E-cadherin和Snail mRNA表达,分析两者在HCC中的表达意义及相关性.结果 E-cadherin mRNA在HCC中的表达水平明显低于癌旁组织(0.41±0.18比1.00±0.14,P<0.01),而Snail mRNA在HCC中的表达水平明显高于癌旁组织(1.78±0.34比1.00±0.25,P<0.01);两者均与HCC的肝内转移、包膜是否完整、血管侵犯及分化程度密切相关(P<0.05);两者在HCC中的表达呈负相关(r=-0.835,P<0.01).结论 E-cadherin和Snail与HCC的侵袭和转移密切相关,可能在HCC侵袭和转移中发挥重要作用.

Objective To detect the expression of E-cadherin and Snail in hepatocellular carcinoma (HCC) and investigate their roles in HCC genesis and development.Methods The expression levels of E-cadherin and Snail mRNA were detected by using real-time quantitative polymerase chain reaction (Real-time PCR) in 36 pairs of HCC and adjacent non-cancerous tissues.The significance and correlation of them in HCC were analyzed.Results E-cadherin mRNA expression was significantly down-regulated in HCC tissues as compared with matched adjacent non-cancerous tissues (0.41 ±0.18 vs.1.00 ± 0.14,P <0.01),and Snail mRNA expression was significantly higher in HCC tissues than in matched adjacent non-cancerous tissues (1.78 ±0.34 vs.1.00 ±0.25,P <0.01).The expression levels of E-cadherin and Snail mRNA were significantly correlated with intrahepatic metastasis,capsular invasion,vascular invasion and tumor grade in HCC (P < 0.05).There was a negative correlation between the expression of E-cadherin and Snail in HCC (r =-0.835,P < 0.01).Conclusion E-cadherin and Snail were significantly correlated with invasion and metastasis of HCC.E-cadherin and Snail likely play important roles in invasion and metastasis of HCC.
