目的 观察降钙素基因相关肽(CGRP)对脑死亡大鼠肺组织水通道蛋白1(AQP-1)及肺水含量的影响.方法 大鼠15只随机分为对照组(A组)、脑死亡组(B组)及CGRP处理组(C组),每组5只.A组持续麻醉维持12h,B组呼吸、循环支持下维持脑死亡状态12h.C组于脑死亡模型建立后静脉输注CGRP 3 μg/kg,随后给予CGRP 0.5μg/(kg·h)持续输注维持12h,维持脑死亡状态12 h.于脑死亡后12h取肺脏组织,测肺系数及肺水含量,苏木素-伊红(HE)染色观察肺脏组织结构变化,逆转录-聚合酶链反应(RT-PCR)方法检测肺组织AQP-1 mRNA水平,Western blot法检测肺组织AQP-1蛋白表达.结果 肺系数及肺水含量B组(7.16 ±0.58、0.91 ±0.07)均较A组(4.12±0.39、0.70±0.08)增高,C组(5.28±0.43、0.80 ±0.05)较A组增高,但较B组低,3组间差异有统计学意义(P<0.05).HE染色A组肺组织结构基本正常;B组出现损伤性改变,C组损伤性改变较B组减轻.肺组织AQP-1 mRNA及蛋白表达水平B组(0.51 ±0.05、0.32±0.05)较A组(0.76±0.03、0.49±0.04)降低,C组(0.98±0.04、0.53 ±0.02)较A、B组增高,组间差异有统计学意义(P<0.05).结论 应用CGRP可提高脑死亡大鼠肺组织AQP-1表达水平,降低肺水含量,减轻肺水肿.

Objective To investigate the pulmonary water content and the mRNAs and the proteins levels of aquaporin-1 (AQP-1) of lung tissues of brain death rats and to investigate the effects of calcitonin gene-related peptide (CGRP) on the Changes of corresponding indicators of brain death rats.Methods 15 adult Wistar rats were randomly grouped into control (A),brain-death (B) and CGRP treatment (C).Brain-death model was established in group B and group C.The group A were placed Foley balloon catheter in intracalvarium only; no brain-death model established.In group C,CGRP 3 μg/kg was injected intravenously immediately after establishment of brain-death model,followed by 0.5 μg/(kg· h)CGRP administered 12 h.At 12 h after the initial brain death,lung tissues were taken,the lung index and lung water content were measured.Histological alterations in lung tissues were examined by hematoxylin and eosin (HE) staining.The levels of AQP-1 were tested by reverse transcriptase-polymerase chain reaction (RT-PCR) and Western blottng.Results In contrast to group A (4.12 ± 0.39,0.70 ± 0.08),lung index and lung water content were higher in group B (7.16 ± 0.58,0.91 ± 0.07) and C (5.28 ± 0.43,0.80 ± 0.05).In contrast to group B,lung index and lung water content were lower in group C (P <0.05).The lung injury was identified by HE staining in group B and C,though less pathologic alterations were shown in group C.The lung Histological examination in group A was normal.The mRNA level and the protein level of AQP-1 in group B (0.51 ± 0.05,0.32 ± 0.05)were lower than that in group A (0.76 ±0.03,0.49 ±0.04),while that in group C (0.98 ±0.04,0.53 ±0.02)were higher than that in group B(P < 0.05).Conclusion CGRP can increase the mRNA level and the protein level of AQP-1,reduce lung water content,and alleviate pulmonary edema in lung tissue of brain death rats.
