目的 探讨pcDNA3.1-血管内皮生长因子(VEGF)165重组质粒转染兔骨髓间充质干细胞(BMSCs),复合聚乳酸/羟基磷灰石(PLA/HA)支架植入股骨头坏死兔体内,观察其治疗效果.方法 逆转录-聚合酶链反应(RT-PCR)检测pcDNA3.1-VEGF165重组质粒转染兔BMSCs后血管内皮细胞生长因子(VEGF)表达.实验分为4组:(1)重组质粒转染BMSCs复合支架组;(2)空白载体转染BMSCs复合支架组;(3)单纯支架植入组;(4)未处理组.分别于术后8、12周对股骨头行显微CT及苏木素-伊红(HE)染色检查,第12周行X线摄影.结果 pcDNA3.1-VEGF165重组质粒转染兔BMSCs后VEGF表达水平明显高于其他组,X线及显微CT可见坏死骨小梁周围部分新骨及小血管形成,其他组可见部分股骨头变形,软骨下骨变薄,骨小梁稀疏、断裂,骨密度不均匀.结论 pcDNA3.1-VEGF165重组质粒转染兔BMSCs复合PLA/HA多孔支架能够诱导兔股骨头内新生血管和骨的形成.

목적 탐토pcDNA3.1-혈관내피생장인자(VEGF)165중조질립전염토골수간충질간세포(BMSCs),복합취유산/간기린회석(PLA/HA)지가식입고골두배사토체내,관찰기치료효과.방법 역전록-취합매련반응(RT-PCR)검측pcDNA3.1-VEGF165중조질립전염토BMSCs후혈관내피세포생장인자(VEGF)표체.실험분위4조:(1)중조질립전염BMSCs복합지가조;(2)공백재체전염BMSCs복합지가조;(3)단순지가식입조;(4)미처리조.분별우술후8、12주대고골두행현미CT급소목소-이홍(HE)염색검사,제12주행X선섭영.결과 pcDNA3.1-VEGF165중조질립전염토BMSCs후VEGF표체수평명현고우기타조,X선급현미CT가견배사골소량주위부분신골급소혈관형성,기타조가견부분고골두변형,연골하골변박,골소량희소、단렬,골밀도불균균.결론 pcDNA3.1-VEGF165중조질립전염토BMSCs복합PLA/HA다공지가능구유도토고골두내신생혈관화골적형성.
Objective To observe the curative effectiveness of implanting the polylactic acid/hydroxyapatite (PLA/HA) porous scaffold combined with rabbit bone marrow mesenchymal stem cells (BMSCs) transfected by plasmid pcDNA3.1-VEGF165 into the rabbit femoral head necrosis model.Methods The expression of vascular endothelial growth factor was detected by using reverse transcriptase-polymerase chain reaction (RT-PCR) after rabbit BMSCs were transfected with pcDNA3.1-VEGF165 plasmid.Four groups were created:pcDNA3.1-VEGF165 plasmid transfection with scaffold group,blank vector transfection with scaffold group,simple scaffold group,and untreated group.Porous scaffolds were implanted into the femoral head.Micro-CT and histological evaluations were performed to observe the rabbit femoral head 8 and 12 weeks after operation.X-ray photography was performed after 12 weeks.Results VEGF expression level was significantly higher in pcDNA3.1-VEGF165 transfection group than other groups.Micro-CT and X-ray showed new bone formation and small blood vessels around the necrotic trabeculae.Femoral head deformation,thin subchondral bone,trabecular bone fracture and non-uniform bone mineral density were shown in other groups.Conclusion Plasmid pcDNA3.1-VEGF165 transfection of rabbit BMSCs with the PLA/HA porous scaffold could induce the formation of blood vessels and bone in rabbit femoral head.
