目的 观察肝素对SD大鼠下切牙颈环区组织骨保护素(OPG)水平及其细胞凋亡的影响.方法 24只4周龄SD大鼠随机分2组,实验组腹部皮下每天注射肝素1 U/g,对照组注射等体积生理盐水.4周后测量股骨骨密度值;下颌骨用微波-乙二胺四乙酸(EDTA)法脱钙切片,行苏木素-伊红(HE)染色、OPG免疫组织化学染色及原位末端转移酶标记(TUNEL)染色,并用Image软件半定量分析OPG含量;透射电镜观察下切牙颈环区细胞超微结构.结果 实验组骨密度值为(0.185±0.006) g/cm2,低于对照组的(0.196 ±0.011) g/cm2 (P <0.05).实验组和对照组下切牙OPG吸光度值分别为0.235 3±0.0350和0.2673 ±0.0444 (P >0.05).TUNEL染色显示实验组颈环区组织未见细胞凋亡,对照组可见个别成牙本质细胞及中间层细胞呈阳性染色.透射电镜见实验组颈环区组织无细胞凋亡发生,对照组有细胞呈现凋亡特征.结论 肝素在诱导SD大鼠骨质疏松的同时,可抑制下切牙颈环区组织细胞凋亡的发生,但OPG水平未见明显改变.

Objective To investigate the effect of heparin on osteoprotegerin (OPG) content and cells apoptosis in the rat cervical loop zone of rat lower incisors.Methods Twenty-four Sprague-Dawley (SD) rats aged 4 weeks were divided into two groups randomly.The experimental group was subcutaneously injected with heparin at the abdomen per day by the dose of 1 U/g,and the control group was injected with normal sodium (NS) by the same volume.All of the rats were sacrificed after 4 weeks,their right femurs were obtained for measurement of the bone mineral density (BMD),and the mandibles were harvested,decalcified and made into paraffin sections,which would be dealt with hematoxylin and eosin (HE)staining,OPG immunohistochemical staining,and TdT-mediated dUTP nick end labeling (TUNEL) staining.The professional software Image Pro Plus was used to semi-quantitatively analyze the content of OPG.The lower incisor cerv;ical loops were separated and prepared to be observed by transmission electron microscopy (TEM).Results The right femur BMD [(0.185 ±0.006) g/cm2] in experimental group was lower than that in control group [(0.196 ±0.011) g/cm2,P <0.05].The Absorbance (A) value of OPG was 0.235 3 ± 0.035 0 in the experimental group and 0.267 3 ± 0.044 4 in control group (P > 0.05).A few odontoblasts and stratum intermedium cells in control group were apoptosis-positive by TUNEL staining;in contrast,there was no apoptosis in the experimental group.The results of TEM examination were similar with TUNEL staining.Conclusion Heparin could lead to osteoporosis and inhibit the apoptosis of odontogenic cells in the rat lower incisor.However,heparin has no significant effect on the OPG content.
