目的 应用基因芯片技术检测颞叶癫痫大鼠与正常大鼠海马的基因表达差异,从基因水平探讨癫痫的发病机制.方法 应用立体定向技术建立大鼠颞叶癫痫模型,行电生理及超微结构检测.提取大鼠海马RNA,进行基因芯片杂交,获取芯片数据.对数据进行比较分析,获取差异表达的基因.按照基因功能分类方法,将差异基因进行功能分类.结果 癫痫大鼠模型的脑电及超微结构出现相应的异常改变.按癫痫组与对照组的基因转录产物表达比值>1.50或<0.67并且统计量>2或<-2为标准筛选,癫痫模型大鼠与正常大鼠比较,差异表达基因113个,其中上调基因46个,下调基因67个.这些差异基因涉及分子功能、生物学进程以及分子组成等功能.结论 基因芯片技术可筛选出癫痫大鼠模型制备前后的差异基因.对差异表达的基因进行分类,可为研究基因功能和传导通路提供基础,为癫痫的治疗提供指导方向.

Objective Differential gene expression was investigated with affymetrix genechip expression analysis between temporal lobe epileptic rat and normal rat.Mechanism of epileptic genesis was investigated from gene level.Methods The temporal lobe epileptic rats were established with stereotactic injection with kainic acid (KA) and tested using electrophysiological and ultrastructural methods.Total RNA samples were isolated from hippocampus.After hybridization,washing and staining,the probe arraies were scanned.The data was analyzed to obtain the differentiated genes including up-regulated and downregulated.According to the method of gene function categorizing,the differentiated genes were categorized into different functions.Results The electroencephalography and ultrastructure of the rat model were abnormal.Compared with normal rat,113 common genes were differentially expressed including 46 genes upregulated and 67 down-regulated in temporal lobe epileptic rat.The genes were categorized into different functions such as molecular function,biological process,cellular component etc.Conclusion The different genes between epileptic rat and normal rat can be obtained from Genechip expression analysis.The genes could be categorized into different functions.The mechanism of action about epileptic genesis based on these experiential data of gene ontology and pathway could be explored from gene level,which provides a potential guiding direction for the treatment of epilepsy.
