目的 观察铜绿假单胞菌-甘露糖敏感血凝菌毛株(PA-MSHA)对膀胱癌细胞株的作用,探讨其作用机制.方法 使用细胞计数试剂盒(CCK-8)法检测肿瘤细胞的增殖,荧光染色法检测细胞凋亡形态,分光光度法检测凋亡因子半胱氨酰天冬氨酸特异性蛋白酶(Caspase)-2和Caspase-9的水平.结果 磷酸盐缓冲液(PBS)组作用T24和5637细胞A450值分别为1.31和1.62,PA-MSHA组A450值分别为0.28和0.41,两组差异有统计学意义(P<0.05);PA-MSHA处理后的膀胱癌细胞出现凋亡小体;PA-MSHA能够活化T24和5637细胞中Caspase-3及Caspase-9蛋白,与PBS组比较差异有统计学意义(P<0.05).结论 PA-MSHA可以抑制T24和5637细胞增殖,并诱导凋亡.

Objective To investigate the effects of pseudomonas aeruginosa-manno sesensitive hemagglutinin (PA-MSHA) on bladder cancer cells and the mechanism.Methods Cell counting kit-8 (CCK-8) assay was used to measure proliferation of tumor cells.The morphologic changes of apoptotic cells were observed by using immunofluorescence staining.Spectrophotometry was used for detection of Caspase-2 and Caspase-9.Results A450 values on T24 and 5637 cells treated with phosphate buffered saline were 1.31 and 1.62,respectively,and those were 0.28 and 0.41 in PA-MSHA group.As compared with cells treated with phosphate buffer (PBS),5637 or T24 cells treated with PA-MSHA for 24 h exhibited much less cytoplasmic organelles.PA-MSHA could induce activation of Caspase-3 and Caspase-9 in T24 cells and 5637 cells.Conclusion PA-MSHA can inhibit proliferation of T24 and 5637 cells,and induce apoptosis.
