目的 观察盐酸氨基葡萄糖(OTL)联合姜黄素对兔膝骨关节炎软骨细胞凋亡的影响.方法 成功建立兔膝关节炎模型,随机分为空白对照组(A组),模型组(B组),盐酸氨基葡萄糖治疗组(C组),联合治疗组(D组),每组10只,经过4周治疗后,采用原位缺口末端标记法(TUNEL)法检测各组软骨细胞凋亡情况,逆转录-聚合酶链反应(RT-PCR)法和免疫组织化学法检测凋亡相关基因B细胞淋巴瘤/白血病-2(bcl-2)、bcl-2相关X蛋白(bax)、p53的基因和蛋白表达水平.结果与B组比较,C组和D组标本的软骨细胞凋亡率及促凋亡基因bax、p53的基因和蛋白表达水平明显降低(P<0.05),而抗凋亡基因bcl-2的基因和蛋白表达水平明显升高(P<0.05),其中D组的抗凋亡效果较C组更为显著(P<0.05),且抗凋亡基因的mRNA和蛋白表达水平结果一致.结论 盐酸氨基葡萄糖可以抑制骨关节炎诱导的软骨细胞凋亡,有一定的保护作用,而姜黄素可以协同盐酸氨基葡萄糖抑制软骨细胞凋亡,减轻炎症,联合应用效果更为显著.
目的 觀察鹽痠氨基葡萄糖(OTL)聯閤薑黃素對兔膝骨關節炎軟骨細胞凋亡的影響.方法 成功建立兔膝關節炎模型,隨機分為空白對照組(A組),模型組(B組),鹽痠氨基葡萄糖治療組(C組),聯閤治療組(D組),每組10隻,經過4週治療後,採用原位缺口末耑標記法(TUNEL)法檢測各組軟骨細胞凋亡情況,逆轉錄-聚閤酶鏈反應(RT-PCR)法和免疫組織化學法檢測凋亡相關基因B細胞淋巴瘤/白血病-2(bcl-2)、bcl-2相關X蛋白(bax)、p53的基因和蛋白錶達水平.結果與B組比較,C組和D組標本的軟骨細胞凋亡率及促凋亡基因bax、p53的基因和蛋白錶達水平明顯降低(P<0.05),而抗凋亡基因bcl-2的基因和蛋白錶達水平明顯升高(P<0.05),其中D組的抗凋亡效果較C組更為顯著(P<0.05),且抗凋亡基因的mRNA和蛋白錶達水平結果一緻.結論 鹽痠氨基葡萄糖可以抑製骨關節炎誘導的軟骨細胞凋亡,有一定的保護作用,而薑黃素可以協同鹽痠氨基葡萄糖抑製軟骨細胞凋亡,減輕炎癥,聯閤應用效果更為顯著.
목적 관찰염산안기포도당(OTL)연합강황소대토슬골관절염연골세포조망적영향.방법 성공건립토슬관절염모형,수궤분위공백대조조(A조),모형조(B조),염산안기포도당치료조(C조),연합치료조(D조),매조10지,경과4주치료후,채용원위결구말단표기법(TUNEL)법검측각조연골세포조망정황,역전록-취합매련반응(RT-PCR)법화면역조직화학법검측조망상관기인B세포림파류/백혈병-2(bcl-2)、bcl-2상관X단백(bax)、p53적기인화단백표체수평.결과여B조비교,C조화D조표본적연골세포조망솔급촉조망기인bax、p53적기인화단백표체수평명현강저(P<0.05),이항조망기인bcl-2적기인화단백표체수평명현승고(P<0.05),기중D조적항조망효과교C조경위현저(P<0.05),차항조망기인적mRNA화단백표체수평결과일치.결론 염산안기포도당가이억제골관절염유도적연골세포조망,유일정적보호작용,이강황소가이협동염산안기포도당억제연골세포조망,감경염증,연합응용효과경위현저.
Objective To investigate the effect of glucosamine hydrochloride glucosamine hydrochloride capsules (OTL) with curcumin on cell apoptosis in the rabbit knee joint osteoarthritis cartilage.Methods Forty rabbit models of osteoarthritis of the knee were successfully established,and randomly divided into control group (A),model group (group B),glucosamine hydrochloride treatment group (group C),curcumin combined treatment group (group D).After four weeks of treatment,the apoptotic chondrocyte of each group was detected by TdT-mediated dUTP nick end labeling (TUNEL) method,and the expression of mRNA and protein level of bcl-2,bax and p53 was determined by the methods of reverse transcription polymerase chain reaction (RT-PCR) and immunohistochemistry.Results Compared with group B,the level of apoptotic chondrocyte and the expression of bax and p53 at the mRNA and protein in group C and D were significantly decreased (P < 0.05),and the level of bcl-2 were significantly increased (P <0.05).Compared with group C,the level of apoptotic chondrocyte in group D was significantly decreased (P < 0.05).Conclusion Glucosamine hydrochloride plays a protective role in inhibit the apoptosis of chondrocyte induced by cartilage osteoarthritis,and the effects of glucosamine hydrochloride combined with curcumin synergistically inhibited chondrocyte apoptosis,reduce inflammation are much more significantly.
