目的 观察脑转移瘤组织中骨桥蛋白(OPN)和细胞黏附因子CD44v6的表达,探讨两者在脑转移瘤发生发展中的作用及机制.方法 采用逆转录-聚合酶链反应(RT-PCR)与链霉菌抗生物素蛋白-过氧化物酶(SP)免疫组织化学法,检测50例脑转移瘤标本中OPN和CD44v6的表达,并探讨两者之间的相关性.结果 OPN和CD44v6 mRNA在脑转移瘤中的相对表达量分别为0.82±0.05、0.77 ±0.05;在原发肿瘤中的相对表达量为0.75±0.07、0.69±0.04,均显著高于正常脑组织(P<0.05),两者的蛋白表达在脑转移瘤中的阳性率分别为92% (46/50)、88% (44/50),与正常脑组织比较明显增高(P<0.05).OPN和CD44v6 mRNA及蛋白在脑转移瘤中的表达均呈明显正相关(r =0.632,P<0.01;r =0.915,P<0.05).结论 OPN和CD44v6在恶性肿瘤的脑转移形成与发展中发挥重要作用,且两者存在一定的协同关系.
目的 觀察腦轉移瘤組織中骨橋蛋白(OPN)和細胞黏附因子CD44v6的錶達,探討兩者在腦轉移瘤髮生髮展中的作用及機製.方法 採用逆轉錄-聚閤酶鏈反應(RT-PCR)與鏈黴菌抗生物素蛋白-過氧化物酶(SP)免疫組織化學法,檢測50例腦轉移瘤標本中OPN和CD44v6的錶達,併探討兩者之間的相關性.結果 OPN和CD44v6 mRNA在腦轉移瘤中的相對錶達量分彆為0.82±0.05、0.77 ±0.05;在原髮腫瘤中的相對錶達量為0.75±0.07、0.69±0.04,均顯著高于正常腦組織(P<0.05),兩者的蛋白錶達在腦轉移瘤中的暘性率分彆為92% (46/50)、88% (44/50),與正常腦組織比較明顯增高(P<0.05).OPN和CD44v6 mRNA及蛋白在腦轉移瘤中的錶達均呈明顯正相關(r =0.632,P<0.01;r =0.915,P<0.05).結論 OPN和CD44v6在噁性腫瘤的腦轉移形成與髮展中髮揮重要作用,且兩者存在一定的協同關繫.
목적 관찰뇌전이류조직중골교단백(OPN)화세포점부인자CD44v6적표체,탐토량자재뇌전이류발생발전중적작용급궤제.방법 채용역전록-취합매련반응(RT-PCR)여련매균항생물소단백-과양화물매(SP)면역조직화학법,검측50례뇌전이류표본중OPN화CD44v6적표체,병탐토량자지간적상관성.결과 OPN화CD44v6 mRNA재뇌전이류중적상대표체량분별위0.82±0.05、0.77 ±0.05;재원발종류중적상대표체량위0.75±0.07、0.69±0.04,균현저고우정상뇌조직(P<0.05),량자적단백표체재뇌전이류중적양성솔분별위92% (46/50)、88% (44/50),여정상뇌조직비교명현증고(P<0.05).OPN화CD44v6 mRNA급단백재뇌전이류중적표체균정명현정상관(r =0.632,P<0.01;r =0.915,P<0.05).결론 OPN화CD44v6재악성종류적뇌전이형성여발전중발휘중요작용,차량자존재일정적협동관계.
Objective To detect the expression of osteopontin (OPN) and cluster of differentiation 44v6 (CD44v6) in brain metastatic tumors,and to explore their role in the formation and invasion of metastases.Methods By reverse transcriptase-polymerase chain reaction (RT-PCR) and immunohistochemical (SP) techniques,the expression of OPN and CD44v6 mRNA and protein was detected in brain metastatic tumors,primary tumor and normal brain tissue.The correlation of their expression was assayed by using statistical methods.Results The relative expression levels of OPN and CD44v6 mRNA were 0.82 ± 0.05 and 0.77 ± 0.05 respectively in brain metastases,and 0.75 ± 0.07 and 0.69 ± 0.04 in primary tumors,which were obviously higher than those in human normal brain tissue (P < 0.05).The positive expression rate of OPN and CD44v6 protein was 92% (46/50) and 88% (44/50) respectively in brain metastases,which was obviously higher than that in human normal brain tissue too (P < 0.05).The expression of OPN mRNA and protein had a positive correlation with that of CD44v6 in metastases (r =0.632,P < 0.01 ; r =0.915,P < 0.05).Conclusion The over-expression of OPN and CD44v6 played an important biological role in the formation of brain metastatic tumors,and they showed a synergistic effect.
