目的 检测三磷酸腺苷(ATP)结合转运蛋白G超家族成员2(ABCG2)和β-微管蛋白Ⅲ(TUBB3)基因在乳腺癌组织中的表达,探讨ABCG2和TUBB3基因与乳腺癌病理特征的关系.方法 196例浸润性乳腺癌组织标本及同标本正常乳腺组织,采用逆转录-聚合酶链反应(RT-PCR)检测ABCG2和TUBB3基因的表达,分析基因表达与临床病理学特征的关系.结果 196例浸润性乳腺癌组织中ABCG2基因的表达率为58.7%,电泳条带灰度值为0.685±0.126,癌旁正常乳腺组织中的表达率为30.1%,基因电泳条带灰度值为0.271±0.143,差异有统计学意义(P<0.05).TUBB3基因的表达率为67.9%,电泳条带灰度值为0.793 ±0.115,癌旁正常乳腺组织中的表达率为23.0%,基因电泳条带灰度值为0.463 ±0.127,差异有统计学意义(P<0.05).ABCG2基因在肿瘤组织的表达与雌激素受体(ER)、孕激素受体(PR)有关(P<0.05),与人类表皮生长因子受体-2(Her-2)、临床分期、脉管癌栓、腋淋巴结转移、肿瘤大小、细胞核增殖抗原(Ki-67)等无明显相关(P>0.05).TUBB3基因在肿瘤组织的表达与腋淋巴结转移、脉管癌栓、肿瘤大小、组织学分级、临床分期、Ki-67、Her-2有关(P<0.05),与ER、PR等无相关(P>0.05).结论 ABCG2、TUBB3基因的异常表达可能在乳腺癌耐药、发生、发展中发挥作用.

목적 검측삼린산선감(ATP)결합전운단백G초가족성원2(ABCG2)화β-미관단백Ⅲ(TUBB3)기인재유선암조직중적표체,탐토ABCG2화TUBB3기인여유선암병리특정적관계.방법 196례침윤성유선암조직표본급동표본정상유선조직,채용역전록-취합매련반응(RT-PCR)검측ABCG2화TUBB3기인적표체,분석기인표체여림상병이학특정적관계.결과 196례침윤성유선암조직중ABCG2기인적표체솔위58.7%,전영조대회도치위0.685±0.126,암방정상유선조직중적표체솔위30.1%,기인전영조대회도치위0.271±0.143,차이유통계학의의(P<0.05).TUBB3기인적표체솔위67.9%,전영조대회도치위0.793 ±0.115,암방정상유선조직중적표체솔위23.0%,기인전영조대회도치위0.463 ±0.127,차이유통계학의의(P<0.05).ABCG2기인재종류조직적표체여자격소수체(ER)、잉격소수체(PR)유관(P<0.05),여인류표피생장인자수체-2(Her-2)、림상분기、맥관암전、액림파결전이、종류대소、세포핵증식항원(Ki-67)등무명현상관(P>0.05).TUBB3기인재종류조직적표체여액림파결전이、맥관암전、종류대소、조직학분급、림상분기、Ki-67、Her-2유관(P<0.05),여ER、PR등무상관(P>0.05).결론 ABCG2、TUBB3기인적이상표체가능재유선암내약、발생、발전중발휘작용.
Objective To investigate the correlations of ATP-binding cassette superfamily G mem-ber 2 (ABCG2) and β-tubulin-Ⅲ (TUBB3)mRNA expressions with formation and development of breast carcinomas.Methods 196 cases of invasive breast cancer tissue specimens and specimens with normal breast tissue were collected.Reverse transcription-polymerase chain reaction (RT-PCR) was used to detect the expressions of ABCG2 and TUBB3 gene.The relationship between gene expression and clinicopatholog-ical features was analyzed.Results The ABCG2 gene expression rate in 196 samples of invasive breast cancer was 58.7% and the electrophoretic bands grayscale was 0.685 ± 0.126,and those in adjacent normal breast tissue were 30.1% and 0.271 ±0.143,respectively (P <0.05).The TUBB3 gene expression rate was 67.9% and electrophoretic bands grayscale was 0.793 ± 0.115 in the samples of invasive breast cancer,and those in adjacent normal breast tissue were 23.0% and 0.463 ± 0.127,respectively (P <0.05).ABCG2 gene expression in tumor tissue was correlated with estrogen receptor (ER) and progester-one receptor (PR) (P < 0.05),but not with human epidermal growth factor receptor-2 (Her-2),clinical stage,vascular invasion,lymph node metastasis,tumor size and proliferation cell nuclear antigen (Ki-67)(P >0.05).The TUBB3 expression in tumor tissue was correlated with axillary lymph node metastasis,vascular invasion,tumor size,histological grade,clinical stage,Ki-67 and Her-2 (P < 0.05),but not with ER and PR (P > 0.05).Conclusion The abnormal expression of ABCG2 and TUBB3 genes may play important roles in drug resistance and development of breast cancer.
