目的 观察甲基化CpG结合蛋白2(MeCP2)在帕金森病细胞模型中的表达.方法 采用免疫荧光技术、Western blot检测MeCP2、酪氨酸羟化酶(TH)蛋白在帕金森病细胞模型中表达的变化.结果 在正常SH-SY5Y细胞中有MeCP2、TH表达,而在帕金森病细胞模型中,免疫荧光检测结果显示MeCP2、TH的表达降低,Western blot结果进一步证实MeCP2、TH的表达下降,MeCP2/β-肌动蛋白(β-actin)由(86.97 ±4.62)%下降至(22.19±2.85)%,TH/β-actin由(49.92±2.35)%下降至(9.02±0.84)%.结论 MeCP2在帕金森病的发病过程中起重要作用,MeCP2可能参与了帕金森病的发病过程.

목적 관찰갑기화CpG결합단백2(MeCP2)재파금삼병세포모형중적표체.방법 채용면역형광기술、Western blot검측MeCP2、락안산간화매(TH)단백재파금삼병세포모형중표체적변화.결과 재정상SH-SY5Y세포중유MeCP2、TH표체,이재파금삼병세포모형중,면역형광검측결과현시MeCP2、TH적표체강저,Western blot결과진일보증실MeCP2、TH적표체하강,MeCP2/β-기동단백(β-actin)유(86.97 ±4.62)%하강지(22.19±2.85)%,TH/β-actin유(49.92±2.35)%하강지(9.02±0.84)%.결론 MeCP2재파금삼병적발병과정중기중요작용,MeCP2가능삼여료파금삼병적발병과정.
Objective To investigate the regularity of methyl-CpG-binding protein 2 (MeCP2) ex-pression in cell model of Parkinson' s disease.Methods Detected the expression of MeCP2 and tyrosine hydroxylase (TH) in cell model of Parkinson' s disease by immunofluorescence and Western blotting.Resuits We found the MeCP2/TH protein expression in SH-SY5Y cell by immunofluorescence and Western blotting,and also the MeCP2 protein and TH protein expression reduced in the cell model of Parkinson's disease.The expression of MeCP2 protein and TH protein reduced were further confirmed by Western blotting analysis,the ratio of MeCP2/β-actin from (86.97 ± 4.62)% decreased to (22.19 ± 2.85)%,and the ratio of TH/β-actin from (49.92 ± 2.35) % decreased to (9.02 ± 0.84) %.Conclusion MeCP2 plays an important role in the pathogenesis of Parkinson' s disease,MeCP2 may be involved in the pathogenesis of Parkinson's disease.
