目的 观察低功率超声在增强顺铂对食管鳞癌细胞抗肿瘤作用中的影响.方法 以食管鳞癌EC9706细胞为实验对象,分别利用单纯低功率超声(0.5 W/cm2和1.0 W/cm2)、单纯顺铂(1 μmol/L)、先超声处理再加入顺铂(超声+顺铂)和先加入顺铂再应用超声(顺铂+超声).4种方式干预细胞的生长,细胞计数试剂盒(CCK-8)法检测细胞活力,显微镜下定性观察荧光染色,流式细胞术定量检测细胞凋亡.结果 超声功率分别为0.5W/cm2和1.0W/cm2时,CCK-8法检测细胞活力,与对照组(3.78%、3.97%)比较,低功率超声(14.36%、15.43%)明显降低了肿瘤细胞活力(P<0.05).超声功率为0.5 W/cm2时,顺铂+超声组(cisPt+ us)细胞凋亡率为(67.23±4.38)%,明显高于超声+顺铂组(us+ cisPt)的(75.38±3.98)% (P<0.05).结论 低功率超声可以提高肿瘤细胞对化疗药物的敏感性,抗肿瘤药物和超声序贯干预的先后顺序,有助于提高肿瘤杀伤疗效.

목적 관찰저공솔초성재증강순박대식관린암세포항종류작용중적영향.방법 이식관린암EC9706세포위실험대상,분별이용단순저공솔초성(0.5 W/cm2화1.0 W/cm2)、단순순박(1 μmol/L)、선초성처리재가입순박(초성+순박)화선가입순박재응용초성(순박+초성).4충방식간예세포적생장,세포계수시제합(CCK-8)법검측세포활력,현미경하정성관찰형광염색,류식세포술정량검측세포조망.결과 초성공솔분별위0.5W/cm2화1.0W/cm2시,CCK-8법검측세포활력,여대조조(3.78%、3.97%)비교,저공솔초성(14.36%、15.43%)명현강저료종류세포활력(P<0.05).초성공솔위0.5 W/cm2시,순박+초성조(cisPt+ us)세포조망솔위(67.23±4.38)%,명현고우초성+순박조(us+ cisPt)적(75.38±3.98)% (P<0.05).결론 저공솔초성가이제고종류세포대화료약물적민감성,항종류약물화초성서관간예적선후순서,유조우제고종류살상료효.
Objective To explore the antitumor effect of cisplatin combined with low-frequency ultrasound on human esophageal squamous cell carcinoma in vitro.Methods Four modes of treatment were used:exposure to low-power ultrasound,application of cisplatin,exposure to ultrasound followed by cispla-tin,and presence of cisplatin followed by exposure to application ultrasound.Ultrasound was used at inten-sities of 0.5 W/cm2 and 1.0 W/cm2 for 10 min,and cisplatin was applied at concentration of 1 μ mol/L per cell suspension treated in EC9706 cells,respectively.The results of each experimental treatment were assessed by cell counting kit-8 (CCK-8) assay.Flow cytometry was performed to examine the apoptosis rate of cells treated with Annexin V-fluoresceine isothiocyanate (FITC)/proliferation index (PI) double staining.Results The combined effect of ultrasound and cisplatin was more effective than that of ultrasound or cisplatin alone.It also appeared that the order of application played a role,with the cisplatin-ul-trasound treatment lowering cell viability more than the ultrasound-cisplatin treatment.Conclusion Low-frequency ultrasound can promote the cytotoxicity of cisplatin and the order of application played a role in enhancing the sensitivity of chemotherapy drugs on tumor cells.
