目的 应用差异蛋白质组学方法筛选结直肠癌组织和对应正常组织的差异蛋白表达.方法 采用双向凝胶电泳(2-DE)分离23例结直肠癌患者和对应正常黏膜组织的总蛋白,从中选取差异表达蛋白质位点,通过基质辅助激光解吸电离飞行时间串联质谱(MALDI-TOF-TOF-MS)鉴定差异表达的蛋白质;然后采用Western blot验证蛋白质组学分析的结果.结果 通过2-DE图谱分析发现8个差异蛋白质斑点,质谱鉴定出6种蛋白质,在结直肠癌患者组织中高表达的有3个,其余3个在正常组织黏膜中低表达.Western blot对表达增高的翻译调节的肿瘤蛋白(TCTP)进行检测,结果与蛋白质组学分析结果一致.结论 蛋白质组学研究是筛选结直肠癌组织中特异性标志物的有效手段.

Objective To screen the tissue proteins associated with the eolorectal carcinoma using a comparative proteomic approach.Methods Two-dimensional electrophoresis was applied to separate the proteins in colorectal cancer specimens and paired normal group from 23 colorectal cancer patients.The proteins with statistically significant differential expression between the colorectal carcinoma and normal groups were identified by matrix-assisted laser desorption/ionization tandem time-of-flight mass spectrometry (MALDI-TOF-TOF-MS).Western blotting was performed to examine the differential expression of the candidate proteins.Results Eight protein spots were found as differentially expressed proteins,in which 6 proteins were identified successfully.Among the identified proteins,there were 3 over-expressed and 3 under-expressed proteins in colorectal cancer tissues as compared with normal group.Translationally controlled tumor protein (TCTP) was found to be over-expressed in colorectal cancer tissues in comparison with the normal group,as confirmed by Western blotting.Conclusion Comparative proteomics analysis provides an effective approach to screen the special biological markers for colorectal cancer patients.
