目的 研究脂肪酸合酶(FASN)在肝癌细胞中的表达及其功能.方法 (1)采用免疫组织化学染色检测肝癌组织中FASN表达,采用实时定量聚合酶链反应(Real-time PCR)及Western blot方法,检测正常肝细胞及不同肝癌细胞(HepG2、SMMC-7721、MHCC97L、MHCC97H、SK-Hep-1)中FASN表达.(2)构建FASN ShRNA真核表达载体,干扰肝癌细胞SK-Hep-1中FASN表达后,检测对肝癌细胞增殖、凋亡及迁移侵袭的影响.结果 (1)与癌旁组织比较,FASN在肝癌组织呈高表达(P<0.05);与正常肝细胞系7702比较,FASN在肝癌细胞系均呈高表达,高转移性的肝癌细胞系MHCC97H与SK-Hep-1中FASN表达高于较低转移性的肝癌细胞系(P<0.05).(2)干扰肝癌细胞SK-Hep-1中FASN后,细胞增殖、迁移和侵袭能力均受到抑制,而细胞凋亡不受影响.结论 (1)FASN在肝癌组织及肝癌细胞系中高表达,且高转移肝癌细胞系表达高于低转移肝癌细胞系;(2)FASN促进了肝癌细胞的增殖,侵袭和迁移,提示FASN可能与肝癌的发生及转移相关.

목적 연구지방산합매(FASN)재간암세포중적표체급기공능.방법 (1)채용면역조직화학염색검측간암조직중FASN표체,채용실시정량취합매련반응(Real-time PCR)급Western blot방법,검측정상간세포급불동간암세포(HepG2、SMMC-7721、MHCC97L、MHCC97H、SK-Hep-1)중FASN표체.(2)구건FASN ShRNA진핵표체재체,간우간암세포SK-Hep-1중FASN표체후,검측대간암세포증식、조망급천이침습적영향.결과 (1)여암방조직비교,FASN재간암조직정고표체(P<0.05);여정상간세포계7702비교,FASN재간암세포계균정고표체,고전이성적간암세포계MHCC97H여SK-Hep-1중FASN표체고우교저전이성적간암세포계(P<0.05).(2)간우간암세포SK-Hep-1중FASN후,세포증식、천이화침습능력균수도억제,이세포조망불수영향.결론 (1)FASN재간암조직급간암세포계중고표체,차고전이간암세포계표체고우저전이간암세포계;(2)FASN촉진료간암세포적증식,침습화천이,제시FASN가능여간암적발생급전이상관.
Objective To study the expression of fatty acid synthase (FASN) in the tissue and cells of hepatocellular carcinoma (HCC) and its effects on the biological characteristics of HCC.Methods (1) Immunohistochemical method was used to detect the expression of FASN in HCC tissues.Real-time quantitative polymerase chain reaction (Real-time PCR) and Western blotting were employed to detect the expression level of FASN in normal hepatocyte and different HCC cell lines (HepG2,SMMC-7721,MHCC97L,MHCC97H,SK-Hep-1).(2) A recombinant plasmid of FASN shRNA was constructed and transfected into the HCC cell SK-Hep-1,cell proliferation,apoptosis,migration and invasion in HCC were analyzed.Results (1) FASN was highly expressed in HCC tissues and HCC cell lines than that in the tumor-adjacent tissueand normal liver hepatocyte 7702 (P < 0.05).The expression of FASN in high metastatic cells MHCC97H and SK-Hep-1 was high than other low metastatic cells.(2) When FASN in SK-Hep-1 cell was interfered,the proliferation,migration and invasion of HCC cells were all inhibited,with no effect on apoptosis of HCC cells.Conclusion (1) FASN is overexpressed both in HCC tissues and HCC cell lines.The expression of FASN in high metastatic HCC cells is high than low metastatic cells.(2) FASN is involved in the promotion of cell proliferation,migration and invasion in HCC cells.
