目的 探讨微小RNA(miR)-421对结直肠癌细胞系SW480增殖的影响及其作用机制.方法 分别上调及干扰miR-421在结直肠癌细胞株SW480中的表达,采用细胞计数试剂盒(CCK-8)法检测细胞的增殖能力;同时采用Western blot检测miR-421与叉头框蛋白O4(FOXO4)的关系.结果 miR-421表达上调325.0％后,连续观察4d,CCK-8结果表明细胞的吸光度值分别为0.309、0.471、0.595、0.687,与阴性对照组(0.306、0.409、0.491、0.554)相比,增殖能力明显增强(P＜0.05);而干扰组miR-421表达下调75.0％,细胞的吸光度值分别为0.303、0.332、0.417、0.473,增殖能力明显减弱(P＜0.05).同时,miR-421过表达及干扰能使FOXO4表达相应的减弱及增强.结论 miR-421表达有助于促进结直肠癌细胞的增殖,其作用机制与其调控FOXO4表达相关.

Objective To investigate the effect of microRNA-421 (miR-421) on the proliferation of SW480 cells and the mechanism.Methods The expression of miR-421 in colorectal cancer cell line SW480 was interfered and upregulated,respectively,then cell counting kit-8 (CCK-8) assay was used to detect the proliferation ability.Meanwhile,Western blotting method was used to evaluate the correlation between miR-421 and forkhead box protein O4 (FOXO4) in SW480 colorectal cell lines.Results After 325.0％ up-regulation of miR-421 in colorectal cancer cell lines SW480,the CCK-8 results showed that the absorbance values for 4 days continuous observation were 0.309、0.471、0.595、0.687 respectively,which was significantly higher than that in normal control (0.306、0.409、0.491、0.554),indicating that the proliferation ability was significantly enhanced (P ＜ 0.05).While the expression of miR-421 was down-regulated to 75％,the absorbance values were 0.303、0.332、0.417、0.473,respectively,with a significant decrease of proliferation (P ＜ 0.05).Meanwhile,the over-expression or down-regulation of miR-421 would significantly decrease or enhance the expression of FOXO4.Conclusion MiR-421 expression can promote the proliferation of colorectal cancer cells,which may be associated with the regulation of FOXO4 expression.
